Isoprenoid alcohols restore protein isoprenylation in a time‐dependent manner independent of protein synthesis

Abstract: Mevalonic acid derivatives are required for the isoprenylation of a variety of growth-regulating proteins. Treatment of NIH3T3 cells with lovastatin (LOV), an HMG-CoA reductase inhibitor, depletes cells of these derivatives and impairs isoprenylation of RAS and RAS-related proteins. In LOV-treated cells, farnesol (FOH) and geranylgeraniol (GGOH) restore RAS and Rap1 isoprenylation, respectively. In this study, we further characterize the manner in which these isoprenoid alcohols are utilized for protein isopre… Show more

“…Therefore, we treated HeLa cells with compactin to block prenylation of Rabs [13] and then incubated cells with geranylgeraniol (GGOH) for different time periods. GGOH was shown to convert to GGPP in the cells [14,15] leading to a subsequent rescue of the prenylation block in the absence of de novo phosphoisoprenoid synthesis and subsequently restores prenylation of Rabs (Figure 5B and Supporting Results S1). Assuming the existence of a Rab prenylation hierarchy, one would expect Rab proteins with higher affinity for REPs to be prenylated faster than the lower affinity Rabs.…”

“…Furthermore, statins reduce the up-regulation of GM1 during T cell activation and prevent the association of Lck and LAT with MR in stimulated T cells. Earlier reports suggested several molecular mechanisms such as an inhibition of HMG CoA reductase (29), prenylation (30), coactivation by integrins like LFA-1 (23), Ag presentation (9), and modulation of lipid microdomains (20,21). The variety of mechanisms might also depend on the used statin concentration.…”

“…Farnesol (FOH) and geranylgeraniol (GGOH) are produced by dephosphorylation of farnesyl pyrophosphate and geranylgeranyl pyrophosphate, respectively, although they are in turn phosphorylated to their pyrophosphates [3-5]. The two isoprenoids are increasingly known to display multiple functions.…”
