Isoporphyrin Intermediate in Heme Oxygenase Catalysis

Abstract: Human heme oxygenase-1 (hHO-1) catalyzes the O 2 -and NADPH-dependent oxidation of heme to biliverdin, CO, and free iron. The first step involves regiospecific insertion of an oxygen atom at the ␣-meso carbon by a ferric hydroperoxide and is predicted to proceed via an isoporphyrin -cation intermediate. Here we report spectroscopic detection of a transient intermediate during oxidation by hHO-1 of ␣-meso-phenylheme-IX, ␣-meso-(p-methylphenyl)-mesoheme-III, and ␣-meso-(p-tri-

“…The β -to-β butadiyne-bridged IP tetramers (23) and meso-tomeso butadiyne-bridged IP tetramers have been studied. The corresponding ZnIP compounds have not been considered because they have a net charge of +4 as each ZnP moiety has a charge of +1.…”

“… 42 Heme oxygenase is an enzyme that catalyzes the degradation of heme and produces biliverdin, iron, and carbon monoxide. 43 45 Biliverdin is subsequently converted to bilirubin by biliverdin reductases . Iron is an essential nutrient required for the survival of most bacteria.…”

“…The aspartic protease studied here is one of the factors involved in decolourisation. To completely decolourise myoglobin and haemoglobin, other enzymes such as oxygenase for haem and oxidoreductase for ferrous ion are probably required. We are currently searching for other enzymes involved in the decolourisation in Aspergillus species.…”
