Isoliquiritigenin Attenuates Adipose Tissue Inflammation and Metabolic Syndrome by Modifying Gut Bacteria Composition in Mice

Abstract: Scope: Isoliquiritigenin (ILG) has been reported to attenuate adipose tissue inflammation and metabolic disorder; however, the underlying mechanisms remain to be elucidated. The aim of this study is to elucidate whether ILG shows the anti-inflammatory and antimetabolic syndrome effects through gut microbiota modification. Methods and results: Mice are fed a high-fat diet (HFD) with or without ILG for up to 12 weeks. The effect of ILG on body weight, blood glucose level, adipose tissue inflammation, gut barrier… Show more

“…For sequencing, 16S rRNA gene sequence libraries were prepared according to the Illumina (San Diego, CA, USA) protocol, as described previously [ 48 ]. The final libraries were pooled, diluted to 4 nM in 5 mM Tris-HCl buffer, and sequenced using a Miseq System with a 600-Cycle Kit (Illumina).…”

“…The Quantitative Insight Into Microbial Ecology 1 (QIIME1) pipeline, an open-source bioinformatics tool for the analysis of raw microbiome DNA sequencing data, was used to investigate the bacterial composition as described previously [ 48 ]. The taxonomy assignment was performed using the EzbioCloud reference database.…”

“…Cecal and fecal SCFAs were measured using GC as previously described. 18 For extraction of cecal and fecal SCFAs, cecal contents and feces were placed in 10 volumes of water and intensively homogenized using a toothpick until fully suspended. The supernatants were transferred to new tubes and heptanoic acid was added as an internal standard.…”

“…For FMT, 100 mg of fresh fecal pellets were collected daily from donor mice immediately after defecation, suspended in 1 mL normal saline, vortexed for 1 min, and centrifuged at 2500 × g for 1 min. Then 100 μL aliquots of supernatant from the stool samples were administered to recipient mice by gavage immediately to minimize the alterations in the composition of fecal microbiome [19].…”
