Actinomycetes are Gram positive bacteria that have been detected in diverse ecological niches. Their member species are known to be a main source of various bioactive compounds. The discovery of Actinomycetes from diverse and unexplored resources has also been linked to increased opportunities to obtain novel bioactive compounds. Insect nest material is being investigated as a new source of novel antimicrobial producing Actinomycetes, which could be harnessed for therapeutic potential. A total of 10 Actinomycetes isolates were collected from the nest of Nasutitermes sp. in Pananjung Pangandaran Nature Reserve. These isolates were evaluated for antimicrobial activity against the challenge bacteria (Eschericia coli, Staphylococcus aureus, Bacillus subtilis, Serratia marcescens) and fungi (Fomitopsis palustris, Fusarium oxysporum, Trichoderma viridae) by dual culture method. The result revealed that several isolates were active against fungi and bacteria. Isolate Pn-TN2 showed the highest level of antibacterial inhibition and the highest antifungal inhibition with Inhibition Rate value more than 80%. By morphological and 16S rRNA gene sequence analysis strongly suggested that the isolate Pn-TN2 belonged to Streptomyces prasinopilosus. We suggested that termite nest is a potential source of bioactive strains of cultivable Actinomycetes for future biotechnological needs. active compounds for further biotechnology application in the medical, pharmaceutical, and agricultural fields.

Endophytic actinomycetes are the potential source for obtaining novel bioactive compounds. Present study evaluates the chemical constituents in Ocimum basillicum and its endophytic actinomycetes. Eleven endophytic actinomycetes were isolated from O. basillicum plant. Among eleven, one isolate was characterized by 16S rRNA and identified as Streptomyces flavoviridis A3WK. A comparative analysis of the FT-IR, GC-MS, antibacterial, antioxidant and anti-inflammatory properties of plant and endophytic actinomycetes extracts was conducted. 12 compounds each of Streptomyces flavoviridis A3WK and O. basillicum were identified by GC-MS. The FT-IR and GC-MS analysis reveals that S. flavoviridis A3WK has more antimicrobial, antioxidant activity and anti-inflammatory properties compared with the results of O. basillicum. Over all the activity of the endophytic actinomycetes was more compared to that of the O. basillicum extracts. The study reveals to show that the existence of endophytic actinomycetes may enhances medicinal value of the medicinal plants, and hence such studies help in the study of endophytic actinomycetes as well as their relevance in host plant. Butanolic extract of soil fungus BioMCC-F.T.3762 fermentation broth exhibited inhibitory activity against Plasmodium falciparum dihydroorotate dehydrogenase (PfDHODH), a fl avin dependent mitochondrial electron transport chain, an essential enzyme in de novo pyrimidine biosynthesis pathway of the parasite. The crude butanolic extract exhibited PfDHODH inhibition of 97% at a concentration of 413 μg/mL, and showed no inhibitory activity against its human homolog. Bioassayguided fractionation was performed on butanolic extract of microbial fermentation broth to isolate the active compounds. The active fraction was isolated using liquid–liquid partition, silica gel column chromatography, octadecylsilane (ODS) column chromatography and was purifi ed by preparative high performance liquid chromatography (HPLC). Major peak of active fractions, FS10-2-12 and FS10-2- 13, were detected by PDA HPLC, which showed a retention time of 12.6 minute and 13.9 minute at 254 nm, respectively. The active fractions exhibited 75% and 40% inhibitory activity against PfDHODH, respectively, at a concentration of 100 μg/mL. In comparison, atpenin A5, a known potent mitochondrial complex II inhibitor exhibited 75% inhibitory activity againts PfDHODH at 366 μg/mL.
