Isolation, purification, and characterization of bovine heart and yeast cytochromes c: An integrated biochemistry laboratory experience

Abstract: This article describes a one-semester project-based laboratory course for biochemistry students centered on the protein cytochrome c. Students learn a variety of protein purification techniques, including ammonium sulfate precipitation, dialysis, ultrafiltration, gel filtration chromatography, ion-exchange chromatography, and electrophoresis, by isolating cytochrome c. The oxidation-reduction properties of this relatively stable and forgiving protein are then characterized by UV-visible spectroscopy and electr… Show more

“…Cytochrome c is a well‐characterized protein which has become a standard used in teaching undergraduate biochemistry students the protein purification procedures and biological assays to fully characterize the protein . A variety of biological assays have been conducted in conjunction with the standard set of cytochrome c protein purification laboratories.…”

“…The biochemistry laboratory at Georgia Southern University uses bovine heart as a source for cytochrome c and includes a preliminary isolation procedure prior to two chromatography steps. The Bradford assay and iron redox assay are followed by SDS‐PAGE (Table ). These experiments include standard protein purification techniques that teach the necessary skills required for anyone interested in pursuing a career using biochemistry .…”

“…As we began to modify our curriculum to focus on relationships between techniques, we searched for an existing project‐based curriculum that could be adapted to our needs. Purification and characterization of a protein seemed to be the obvious choice, and several successful versions do appear in the literature . However, these experiments were difficult to incorporate into our curriculum as written, because they required at minimum 4–6 hours of class time per week and were designed for a class size of 16 students or fewer.…”
