An aqueous two-phase system was developed to extract anthocyanins present in the fruit residue during juice production from the fruit of Vaccinium uliginosum Linn. A maximum partition coefficient of 10.67 and a recovery of 96.09% for anthocyanins could be obtained using an extraction system consisted of 30% (w/w) ethanol and 19% ammonium sulfate. Compared with the traditional extraction using acidified ethanol, the novel aqueous two-phase extraction could not only yield a much higher concentration of anthocyanins, save more ethanol, energy, and time, but also decrease impurities in extract, e.g. proteins and sugars by 58% and 66%, respectively. AB-8 macroporous resin was applied to the purification of anthocyanins. A novel and simple separation technique for anthocyanins was developed by integrating aqueous two-phase extraction and macroporous resin column chromatography. This new technology might be a suitable for other bioactive natural products on industrial scale.

Botanical Background and Cultivation ! Aronia berries originate from the eastern parts of North America and East Canada [1], [2], [3]. Their migration to Europe occurred around 1900 via Germany to Russia and back. Around 1946 the plant was established as a cultivar in the former Soviet Union. More recently it is cultivated also in East European countries and Germany (Oberlausitz) [1], [2]. The genus name Aronia has been replacing the common name, chokeberry. Two species can be distinguished: Aronia melanocarpa [Michx.] Elliot (black chokeberry, Aronia noir) and Aronia arbutifolia [L.] Elliot (red chokeberry, Aronia rouge) [3]. A third entity is intermediate and has variously been considered a species (A. prunifolia), a variety of each of the other two, or a hybrid [3]. The other common names for Aronia besides chokeberries include black apple berry and rowanberries (Sorbus aucuparia L.), whereby in the latter case the hybrids of rowanberry with Aronia are likely to be addressed [4], [5], [6]. Aronia is a member of the Rosaceae family, and the cultivars used for fruit production are from the species Aronia melanocarpa [6], [7], [8], [9]. Cultivars have been bred for larger edible fruits and ornamental value. The plants have no known pest problems [10]. Some cultivars are bred of true black chokeberry (Aronia melanocarpa) and some are hybrid cultivars (e. g., Aronia x Sorbus) [5], [11]. The more important cultivars include "Nero" (Czechia), "Rubina" (crossing from Russian and Finnish plants), "Viking" (Finland), "Kurkumäcki" (Finland), "Hugin" (Sweden), "Fertödi" (Hungary) and "Aron" (Denmark) [2]. "Viking" and "Nero" are cultivars that are commonly available in North American plant catalogues [7]. The Aronia shrubs can grow to a height of 2-3 m, which produce in May to June umbels of some 30 small white flowers, ripening to bright red (red chokeberry) or to purplish black berries (black chokeberry; Ø 6-13 mm, 0.5-2 g) [1], [2], [8], [12]. Harvest is performed mechanically between August and September. Five to twelve tons per hectare can be expected in about five years, once plants have matured [7], [8]. The effect of fertiliser rate on fruit quality parameters in the black chokeberry showed that an increased fertiliser application results in increased growth and higher yield whereas pigment content and total acidity decreased. Maximum production of anthocyanins per plant was obtained with a medium high fertiliser rate (50 kg N/ha, 44 kg P/ha, 100 kg K/ ha) [13]. The content of undesirable brown compounds and that of desirable anthocyanins is also dependent on maturation as documented in [11]. IntroducciónLa ficocianobilina es el pigmento tetrapirrólico que constituye el grupo prostético de la C-ficocianina (CPC) y la Aloficocianina (APC). Estas proteínas son constituyentes de los ficobilisomas presentes en las algas verde-azules 1 y han sido asociadas a propiedades antioxidantes, inmunomoduladoras y citoprotectoras. 2-6Adicionalmente se ha demostrado que muchas de estas propiedades biológicas están asociadas al sistema tetrapirrólico que está presente en estas proteínas como grupo prostético. 2,7,8 Presumiblemente la acción de la PCB está relacionada con la inhibición de la enzima NADPH oxidasa y con su capacidad antioxidante . 2,9,10 Adicionalmente debido a sus propiedades físico-quí-micas es posible que esta molécula pueda ejercer su acción traspasando la membrana hemato-encefálica, por lo que potencialmente podría ser un producto con aplicaciones en el tratamiento de la isquemia cerebral. 7,11 Los métodos descritos para la purificación y caracterización de la PCB implican la extracción previa de las ficobiliproteínas en las que está contenido este grupo prostético. Posteriormente el pigmento es liberado de la estructura polipeptídica mediante el tratamiento con ácido concentrado o reflujo con un alcohol . 12,13 La extracción posterior se realiza con solventes orgáni-cos como cloroformo o diclorometano debido a la solubilidad del pigmento en estos sistemas. Finalmente, el proceso de purificación puede implicar un método cromatográfico ya sea en placa, batch o columna. 14 Una alternativa que emplea herramientas biotecnológicas está asociada a la co-expresión de las enzimas involucradas en la modificación del grupo hemo a Biliverdina (BV) y posteriormente a PCB.9 El organismo RESUMENLa ficocianobilina es el grupo prostético de las ficobiliproteínas C-ficocianina y Aloficocianina, las que se encuentran presentes en las algas verde-azules. Entre las estrategias empleadas para la purificación de este pigmento tetrapirrólico se encuentra la extracción de las proteínas y posterior ruptura del enlace tioéter, mediante el que está unido las cadenas polipeptídicas. Posteriormente se lleva a cabo el proceso de extracción con un solvente orgánico y la purificación basada en la cromatografía en placa, en batch o columna. La caracterización de esta molécula implica métodos cromatográficos, espectroscópicos y la determinación de la actividad antioxidante. En esta revisión presentamos un resumen de las estrategias de purificación empleadas para la obtención de este pigmento, así como los métodos de caracterización fundamentales. Adicionalmente comentamos sobre posibles estrategias que se pudieran emplear para la obtención de preparaciones de la PCB con elevadas purezas, que puedan ser empleadas para la evaluación de sus propiedades potenciales en la industria biofarmacéutica.Palabras clave: ficocianobilina, tetrapirrol, grupo-prostético, ficobiliproteínas.
