Isolation of Trypanosoma Cruzi from the Blood of Infected Mice by Column Chromatography

Abstract: Trypomastogotes of three strains of Trypanosoma cruzi were isolated from the blood of infected mice employing lymphoprep for separation of the red blood cells and a column of DEAE cellulose for removal of white cells and platelets. An average recovery of 45 to 58 percent of actively motile, infective organisms, free of contaminating blood cells was obtained. Protein and carbohydrate assays of the separated organisms revealed significant differences between the Tulahuen, a reticulotropic strain, and the House 5… Show more

“…Trypomastigotes of T. cruzi (Tulahuen isolate) were purified from the blood of Crl-CDl(1CR) Swiss mice (Charles River Laboratories, Portage, MI, USA) infected subcutaneously with 1 x lo6 organisms 10-12 days previously (Budzko & Kierszenbaum 1974, Mercado & Katusha 1979. The organisms were washed with, and resuspended at the desired concentration in RPMI 1640 medium (Gibco, Grand Island, NY, USA) containing 100 IU/ml penicillin, 100 pg/ml streptomycin and 5% heat-inactivated foetal bovine serum (FBS; Hyclone, UT, USA).…”

“…Trypomastigotes were purified 10 to 12 days later from the blood by centrifugation through Isolymph (Gallard-Schlesinger, Carle Place, N.Y.) (5) followed by column chromatography through DEAEcellulose (15). The eluted trypanosomes were washed twice with Dulbecco's modified minimal essential medium (GIBCO, Grand Island, N.Y.) containing 100 IU of penicillin and 100 tg of streptomycin per ml and supplemented with 10% fetal bovine serum (Sigma Chemical Co., St. Louis, * Corresponding author.…”

“…The trypomastigotes were purified by centrifugation over Isolymph (Gallard Schlesinger, Carle Place, NY) (27) followed by chromatography through a DEAE-cellulose column (28). The eluted parasites were concentrated by centrifugation (800 x g, 15 min, 200C) and resuspended at 3-5 x 106 organisms per ml in the desired medium.…”
