Isolation of the chicken middle-molecular weight neurofilament (NF-M) gene and characterization of its promoter

Abstract: We have isolated and sequenced genomic DNA clones covering the coding region of the chicken mid-size neurofilament (NF-M) gene and greater than 1 kb of its 5' upstream region. The NF-M gene contains two introns which both are located within the highly conserved C-terminal region of the rod domain. The 5' end of the corresponding mRNA was assigned to a G residue 40 nucleotides upstream of the translation start site and in appropriate distance from a potential TATA box. To functionally analyze the NF-M promoter,… Show more

“…Significant developmental increases also occur in the modulatory tonus of cholinergic and catecholaminergic inputs and in the expression of brain-derived growth factor (BDNF) in the song system Saito, 1989, 1991;Akutagawa and Konishi, 1998). The expression of both chicken and rat NF-M is upregulated in neuronal cells upon treatment with neurotrophic factors (Verge et al, 1990;Zopf et al, 1990). Moreover, the expression of BDNF in the brain of juvenile zebra finches shows regional and temporal changes similar to those we describe here for NF-M (Akutagawa and Konishi, 1998), raising the possibility that BDNF might be involved in the regulation of NF-M expression.…”

“…This residue probably was not phosphorylated in pep- (Zopf et al, 1990). All phosphopeptide fragments identified to date are indicated below the sequence, including those in the acidic region reported previously (L3-L5, V5-V7; Hollander et al, 1996) and those analyzed in the present report (refer to Fig.…”

“…We have focused on the ana!ysis of chicken NF-M. The deduced sequence of the chicken NF-M polypeptide (Zopf et al, 1990) revea!s the presence of the same motifs found to be phosphory!ated in the rat NF-M: a single KSD and four repeats of a KSP motif. The chicken NF-M protein differs from the mammalian counterparts in that it contains 17 repeats of the KXXS/TP motif, which appears on!y once in human, rat, and mouse NF-M.…”

Monteiro et al (1990) have also generated transgenic mice containing the mouse NF-L gene (pNF-L[Bam]).…”
