Isolation of the amplified dihydrofolate reductase domain from methotrexate-resistant Chinese hamster ovary cells.

Abstract: We isolated overlapping recombinant cosmids that represent the equivalent of two complete dihydrofolate reductase amplicon types from the methotrexate-resistant CHO cell line CHOC400. The type I amplicons are 260 kilobases long, are arranged in head-to-tail fashion, and represent 10 to 15% of the amplicons in the CHOC400 genome. The type II amplicons are 220 kilobases long, are arranged in head-to-head and tail-to-tail configurations, and constituted the majority of the remaining amplicons in CHOC400 cells. Th… Show more

“…The major repeating unit (amplicon) in this cell line is 240 kb in length (6). Results of intrinsic labeling studies on synchronized CHOC 400 cells were consistent with the presence of one (7,8), or possibly two (9,10), fixed initiation sites lying in the intergenic region between the DHFR and 2BE2121 genes [termed ori-f and ori-' (10); Fig.…”

“…In many cases characterized, these amplified regions contain large ''head-to-head'' duplications (palindromes) of sequences that can be tens to hundreds of kb in size (3)(4)(5)(6)(7)(8). Cytogenetic studies have further revealed that large inverted duplications of a chromosomal region are formed at early stages of gene amplification (9)(10)(11).…”

“…Thus, during the amplification process, pieces of DNA from different parts of the genome are joined by recombination to form novel structures and novel joints (new restriction fragments). In some highly resistant cell lines, novel structures are amplified enough to be cloned by differential screening or to be detected by direct visualization or in gel renaturation, and highly redundant novel joints have been detected (2,9,22,32,37). In these cases, most of the amplified units seem to be derived from a single rearranged structure, formed early in * Corresponding author.…”
