ISOLATION OF POLYSOMES AND POLY(A)‐CONTAINING RNA FROM CHLORELLA FUSCA VAR. VACUOLATA

Abstract: SUMMARYRibonuclease activity in 120000 g supernatant extracts of Chlorella fusca var, vacuolata showed maximal activity at pH 5 and minimal activity at about pH 7-5 in the range 5 to 8-6, Dithiothreitol, high salt concentration and 100 mM tris significantly inhibited ribonuclease activity although the extent of inhibition was not uniform at all pH values, Heparin, polyvinylsulphate and sodium dodecylsulphate were largely ineffective as ribonuclease inhibitors under the same conditions, Polysomes were isolated … Show more

“…The mycelial powder and residual solid CO, were transferred to 50 ml polythene tubes (Falcon). After allowing the residual CO, to sublime, but before the mycelial fragments thawed, RNA was extracted using triisopropylnaphthalene sulphonate and phenol/cresol, followed by ethanol precipitation as described previously (Leonard et al, 1981).…”

“…It is thought that a minimum of two units are required for high-affinity heat-shock binding and such arrays are present in both sequences. HSE involvement in control of ligninolytic enzymes has been extensively studied with respect to manganese peroxidases of Phanerochaete chrysosporium (Gold & Alic, 1993;Li et al, 1995). There may be other similarities between manganese peroxidase and laccase gene expression, as both may be controlled by metal response elements (MREs) .…”
