Isolation of Mitochondria from Minimal Quantities of Mouse Skeletal Muscle for High Throughput Microplate Respiratory Measurements

Abstract: Dysfunctional skeletal muscle mitochondria play a role in altered metabolism observed with aging, obesity and Type II diabetes. Mitochondrial respirometric assays from isolated mitochondrial preparations allow for the assessment of mitochondrial function, as well as determination of the mechanism(s) of action of drugs and proteins that modulate metabolism. Current isolation procedures often require large quantities of tissue to yield high quality mitochondria necessary for respirometric assays. The methods pre… Show more

“…To analyze whether the observed changes in OCR could be due to diminished mitochondrial integrity rather than ETC dysfunction, we measured citrate synthase (CS) activity. This has previously been described as a measure of mitochondrial integrity ( Boutagy et al., 2015 ; Short et al., 2005 ). Our findings suggest that LPO treatment has no marked impact on CS activity ( Figure 3 J), suggesting that mitochondria remain intact following intracellular lipid accumulation.…”

“…Mitochondrial isolation from the skeletal muscle was modified from the protocol described by Boutagy [ 30 ]. Briefly, red muscle was removed from the quadriceps muscle and finely minced with scissors.…”

“…Mitochondria were isolated from gastrocnemius muscles, and mitochondrial bioenergetic functions were analyzed as previously described (3,4,19). The oxygen consumption rate (OCR) measurement was performed using a Seahorse XFp Extracellular Flux Analyzer (Seahorse Bioscience, North Billerica, MA) following the manufacturer's instructions.…”
