Isolation of Inhibin α-Subunit Precursor Proteins from Bovine Follicular Fluid*

Abstract: Two proteins with structural characteristics similar to peptide sequences identified in the inhibin alpha-subunit precursor sequence have been isolated from bovine follicular fluid. A side-fraction from the purification of bovine follicular fluid inhibin with high levels of inhibin immunoactivity relative to its inhibin bioactivity was fractionated through a sequence of procedures which included triazine dye affinity and phenyl-Sepharose chromatography, gel permeation chromatography on Sephadex G-100, reverse … Show more

“…Inhibin is mainly produced by granulosa cells and is detectable in follicular fluids with multiple molecular forms in female mammals [3,[22][23][24][25]. After injections of inhibin antiserum or NGS, Ig G enters the follicular fluid of antral follicles in the ovary, and this makes it possible for inhibin antibodies, one kind of Ig G, to combine with inhibins in follicular fluid and have local functions in follicular development.…”

“…The concentrations of inhibin were determined by RIA as previously described (McNatty et al 1992). The antibody to inhibin detects 31 kDa inhibin and the pro-aC subunit (Robertson et al 1989). The intra-and inter-assay coefficients of variations were 9.4% and 13.1% respectively.…”
