Isolation of infectious pancreatic necrosis (IPN) virus from non‐salmonid fish

Abstract: The isolation and charaeterization of infectious pancreatic necrosis (IPN) virus from a goldfish, discus fish and bream is described. The fish from whieh the isolates were recovered showed no pathologieal signs of IPN. All three virus isolates were neutralized by antiserum to IPN, strain Ab, but not by antiserum to the Sp or VR-299 strains. They were morphologically identical to IPN virus in negative stain electron mieroscopy, grew in the eytoplasm of BF-2 cells, as shown by immunofluorescence and, like IPNV, … Show more

“…Cells were grown in MEM with Earle's salts (Flow Laboratories) supplemented with 10 ~ foetal calf serum (MEM-10), penicillin (100 international units/ml), streptomycin (100 ~tg/ml), gentamicin (25 ~tg/ml) and fungizone (0-25 ~tg/ml). Procedures for virus isolation were similar to those described by Adair & Ferguson (1981). Tissue samples from different organs of diseased rainbow trout or eel were homogenized with nine volumes of MEM containing double strength antibiotics.…”

“…The isolated virus was not pathogenic to the original host nor to rainbow trout fry. The IPNV-Ab-subtype has been isolated from rainbow trout UBRGENSEN, 1971); from goldfish, discus fish and bream (ADAIR and FERGUSON, 1981); from southern flounder (MCALLISTER et al, 1983); from Japanese eel and tilapia . The IPNV-Ab-subtyp usually has a low virulence.…”

“…Two of these serotypes, AB and SP (Jorgensen & Kehlet, 1971), are found only in Europe. Both the AB and SP serotypes have been detected in trout, in several nonsalmonid species and in elvers in Europe (Hudson et al, 1981 ;Adair & Ferguson, 1981 ;Castric & Chastel, 1980; Munro et al, 1976; Ahne, 1980). The third serotype of IPNV is prevalent in North America and is represented by reference strain VR-299 (Wolf & Quimby, 1971).…”
