Isolation of Streptomyces Species from Soil

Abstract: This unit describes a general protocol for the isolation of Streptomyces species from soil and fresh water, using a procedure for the selective growth of Streptomyces species. Preparation of the necessary growth medium, recognition of the morphology of the bacteria, and safety considerations are also covered. Curr. Protoc. Microbiol. 19:10E.4.1‐10E.4.5. © 2010 by John Wiley & Sons, Inc.

“…Although the temperature used in this modified method was lower than that in Kharel et al (2010), we managed to isolate Streptomyces strains from the soil samples from King George Island. The isolates exhibited morphometric and ultrastructural characteristics consistent with the Streptomyces genus (Fig.…”

“…1). A modified method described by Kharel et al (2010) was used to isolate Streptomyces species and kill most of the non-spore-forming microbes. One gram of soil sample was weighed and suspended in 10 ml of sterile distilled water.…”

“…Streptomyces are Gram-positive bacteria and are the largest representative genus of actinomycetes, which are found mostly in soil 34 . They are industrially useful microorganisms that produce a wide variety of antibiotics, and several studies have been carried to investigate the biological effects of Streptomyces species isolated from soil samples 35,36,37 .…”

“…Experiment 3 could be completed using a different array of bacterial isolates from public, private, or student-generated (depending on class time) culture collections. The bacterial cultures used in the field test of this module utilized Streptomyces species obtained from outdoor air (Weber et al, unpublished data), but this genus can be isolated from soils by using methods described by Kharel et al ( 12 ).…”
