Isolation of glycopeptides with skin test activity from dermatophytes

Abstract: By using ethylene glycol extraction of whole submerged cultures, followed by Sephadex G-200 and diethylaminoethyl-Sephadex chromatography, we isolated four distinct glycopeptides from Trichophyton mentagrophytes, T. rubrum, and Microsporum canis. Chemical analyses revealed that these glycopeptides contained mostly carbohydrate (42.5 to 81.6%) and protein (4.3 to 11.3%), with lesser amounts of phosphorus (0.4 to 6.0%) and hexosamines (0.3 to 0.6%). Based upon total carbohydrate and monosaccharide content, these… Show more

“…Evidence suggests that a carbohydrate/protein complex mediates both humoral and cell-mediated immunity (10,26). Moser and Pollack have isolated a series of glycopeptides from dermatophytes which were capable of inducing delayed hypersensitivity in guinea pigs (26). Both cross-reactive and species-specific antigen fractions were identified.…”

“…Portiotis of this work were reported at the Society for Investigative Dertnatology Annual Meeting in Washington D.C. on April [26][27][28][29][30]1989. An abstract of this presentation appears in (Tlin Res 37: 652A, 1989. quinekeanum in sublethally irradiated BALB/c mice as measured by the size of the fungal lesion (32).…”

“…The dermatophytes, like other fungi, have a very complicated antigenic makeup. Antigenic substances of the dermatophytes may be glycopeptides, peptides, or carbohydrates, and the individual types of antigens may elicit different types of responses (14,184). In addition, the keratinase of Trichophyton mentagrophytes may be an important antigen not only because it elicits strong delayed hypersensitivity responses but also because antibodies that inhibit the proteolytic activity of this enzyme are produced (97).…”
