Isolation of Glycoalkaloids with the Chromatotron and Their Determination by High-Performance Liquid Chromatography

Abstract: An e x t r a c t of Solanum l a c i n i a t u m leaves was f r a c t i o n a t e d w i t h t h e Chromatotron. The m k a l o i d f r a c t i o n t h u s o b t a i n e d was analyzed by reversed-phase HPLC. The l o a d c a p a c i t y o f t h e Chromatotron w i t h a 2-mm l a y e r t h i c k n e s s i s 600 mg (as determined w i t h c h o l e s t e r o l ) and t h e recovery o f solasonine from t h e p l a n t e x t r a c t (as determined by HPLC) i s 93-96%. “…The crude glycoalkaloids can be precipitated by the addition of NH4 0H; extraction of the supernatant with butanol yields the saponin fraction. The direct identification of glycoalkaloids (e.g., solasonine, solamargine) is possible by HPLC (Elridge and Hockridge 1983;Hunter and Heftmann 1983; in combination with fast atom bombardment (FAB) or thermospray mass spectrometry (Price et al 1985;Ehmke et al 1987). For the sensitive and specific determination of solasodine glycosides in minute samples, Weiler et al (1980) established a radioimmunoassay.…”
