In this study, chromosome painting was developed and used to identify alien chromosomes in TAi-27, a wheat - Thinopyrum intermedium addition line, and the chromosomes of the three different genomes of Th. Intermedium. The smallest alien chromosome of TAi-27 was microdissected and its DNA amplified by DOP-PCR was used as a probe to hybridize with metaphase chromosomes of TAi-27 and Th . intermedium . Results showed that hybridization signals were observed in all regions of a pair of the smallest alien chromosomes and the pericentromeric area of another pair of alien chromosomes in TAi-27, indicating that the probe from microdissected chromosome is species specific. In Th . intermedium , 14 chromosomes had wide and strong hybridization signals distributed mainly on the pericentromere area and 9 chromosomes with narrow and weak signals on the pericentromere area. The remaining chromosomes displayed a very weak or no signal. Sequential FISH/GISH on Th . intermedium chromosomes using the DNAs of microdissected chromosome, Pseudoroegneria spicata (St genome) and pDbH12 (a Js genome specific probe) as the probes indicated that the microdissected chromosome belonged to the St genome, three genomes (Js, J and St) in Th . intermedium could be distinguished, in which there is no hybridization signal on J genome that is similar to the genome of Th . bessarabicum . Our results showed that the smallest alien chromosomes may represent a truncated chromosome and the repetitive sequence distribution might be similar in different chromosomes within the St genome. However, the repetitive sequence distributions are different within the Js genome, within a single chromosome, and among different genomes in Th . intermedium . Our results suggested that chromosome painting could be feasible in some plants and useful in detecting chromosome variation and repetitive sequence distribution in different genomes of polyploidy plants, which is helpful for understanding the evolution of different genomes in polyploid plants.

Agropyron cristatum (L.) Gaertn. (2n = 4x = 28, PPPP), a wild relative of wheat, could provide many desirable genes for wheat improvement. Microdissection and degenerate oligonucleotide primed –polymerase chain reaction (DOP–PCR) is an effective way to isolate specific sequences. The development of specific sequences and functional markers of P genome could lay the foundation for gene mapping and cloning. In this study, chromosomes 6PS and 6PL were microdissected from wheat–A. cristatum 6PS and 6PL addition lines, and DOP–PCR products from both microdissected chromosomes were used as probes to hybridize with mitotic metaphase chromosomes of diploid A. cristatum, wheat–A. cristatum addition lines 6PS, 6PL, and 6P. The results showed that the distribution patterns on A. cristatum chromosomes using DOP–PCR products of microdissected 6PS as the probe were similar to those using DOP–PCR products of microdissected 6PL as the probe. They both distributed along all chromosomes, and chromosome 6P and other chromosomes were similar according to the hybridization pattern. There were no signals on wheat chromosomes indicating that DOP–PCR products contain P‐genome specific sequences. Actually, we obtained a P‐genome specific sequence from the DOP–PCR products of 6PS, which could be used as a fluoresence in situ hybridization (FISH) probe to identify P genome chromatin in wheat–A. cristatum introgression lines. These results could provide a basis for the study of structure and evolution of P genome chromosomes. Furthermore, this study may lead to mapping and cloning of the desirable genes on 6P chromosome.

Resistance to Wheat streak mosaic virus– a survey of resources and development of molecular markers

Wheat streak mosaic virus (WSMV) has been newly documented in Australia. The vulnerability of contemporary Australian elite wheat germplasm prompted a survey for effective resistance against an Australian isolate, WSMV-ACT. This study confirms the effectiveness of previously reported sources of resistance and shows that new sources of resistance also confer protection. The resistance derived from Thinopyrum intermedium (Wsm1) as a 4D translocation and a new 4A translocation, and two bread wheat resistances, Wsm2 and the new source c2652, were effective against WSMV-ACT in glasshouse experiments. Wsm1 was effective at lower temperatures but ineffective above 20°C, a temperature sensitivity shared with many of the derivatives of Wsm2 except for one new selection which was effective at 26°C. True wheats c2652 and Wsm2 selection CA745, and amphiploids Zhong1, Zhong2, Zhong4, Zhong5, TAF46, Summer1, Ot38 and OK7211542 were uniformly resistant at 20, 25 and 28°C. New sources of resistance were identified in a Th. scirpeum-wheat amphiploid, B84-994, and in chromosome addition lines Z2, Z6 and TAi27, derived from wheat-Th. intermedium partial amphiploids. Several new, tightly linked SSR, RAPD and EST-ILP PCR markers were developed for tracking the various Th. intermedium translocations associated with Wsm1, including the smaller translocations on wheat chromosome 4AS and 4DS. Three markers for the 4A-Wsm1 translocation were validated on a segregating breeding population.
