Isolation of chicken taste buds for real‐time Ca2+ imaging

Abstract: We isolated chicken taste buds and used a real-time Ca2+ imaging technique to investigate the functions of the taste cells. With RT-PCR, we found that isolated chicken taste bud-like cell subsets express chicken gustducin messenger RNA. Immunocytochemical techniques revealed that the cell subsets were also immunopositive for chicken gustducin. These results provided strong evidence that the isolated cell subsets contain chicken taste buds. The isolated cell subsets were spindle-shaped and approximately 61-75 μ… Show more

“…In rats, taste bud cells that express α-Gustducin have a long life span compared to other cell types [27]. α -Gustducin is explicitly expressed in mature chicken taste buds at P3 [9] and is highly expressed in an isolated chicken taste bud-like subset of cells [30]. In the present study, we detected α-Gustducin immunosignals in immature taste buds as early as E18.…”

“…Chickens show aversive responses to bitter and sour (Ganchrow et al 1990;Kudo et al 2010a;Hirose et al 2015;Dey et al 2017) and a preference for feeds containing fat and umami tastants (Sawamura et al 2015;Yoshida et al 2015), and we demonstrated that isolated taste bud cells of chickens responded to umami and bitter solutions (Kudo et al 2014). These findings indicate that the taste sense of chickens is similar to that of mammals.…”

“…It has been reported that α-Gustducin is specifically expressed in chicken taste buds911. Two molecular markers, Vimentin and α-Gustducin, were used to label taste buds in the epithelial sheets of the base of the oral cavity in P0 to P8 chickens.…”
