Isolation of cDNA clones of genes with altered expression levels in phosphate-starved Brassica nigra suspension cells

Abstract: Differential gene expression at the transcriptional level was examined as an initial step in the investigation of the P(i) starvation response of Brassica nigra suspension cells. Total RNA was extracted from 7-day old cells grown in media containing either no P(i), 1.25 mM or 10 mM Pi. In vitro translation was carried out using their respective poly(A)+ RNA isolates and the resultant polypeptides were separated on a high-resolution SDS-PAGE gel. Scanning densitometry identified four polypeptides (ca. 31.7, 32.… Show more

“…Several experiments have studied the role of high-affinity P-transporters in plant P uptake. Malboobi and Lefebvre (1995) and Leggewie et al (1997) both reported higher transcript levels for the high-affinity P-transporter under conditions of P deprivation. Muchhal and Raghothama (1999) reported that the expression of the P-transporter, LePT1, in tomato was altered by P availability of the growth medium.…”

“…Differential Transcriptional Responses of Arabidopsis PAPs to Phosphate Deprivation-Because suspension cultures of different plant species had been employed in past studies on the regulation of many types of genes by phosphate deprivation treatment (31,(45)(46)(47)(48)(49)(50)(51)(52)(53), the transcriptional responses of AtPAP7-AtPAP13 to phosphate deprivation treatment were investigated using Arabidopsis suspension cells. Total RNA was prepared from both control and phosphate-deprived cell samples.…”
