Isolation of Bacillus sp. A5.3 Strain with Keratinolytic Activity

Abstract: Environmental safety and economic factors necessitate a search for new ways of processing poultry farm feathers, which are 90% β-keratin and can be used as a cheap source of amino acids and peptones. In this study, feather-decomposing bacteria were isolated from a site of accumulation of rotten feathers and identified as Bacillus. Among them, the Bacillus sp. A5.3 isolate showed the best keratinolytic properties. Scanning electron microscopy indicated that Bacillus sp. A5.3 cells closely adhere to the feather … Show more

“…We do not have a clear explanation for the high thermal stability of myrosinase under this investigation which is higher than that reported so far for other microbial myrosinases, even those purified from fungi (Rakariyatham et al 2005 ). However, it can be attributed to the nature of its source, as the genus Bacillus is known as a potential source of producing thermostable enzymes (Cotta et al 2021 ; Aktayeva et al 2022 ). Additionally, the myrosinase in this study appears to prefer alkaline conditions (with an optimal pH of 8.00) compared to myrosinases previously isolated from other bacteria that had optimal pH values in the range of 6.0–6.6 (Tie et al 2021 ; Cebeci et al 2022 ).…”

“…Strains were identified by proteomic profiling of ribosomal proteins and by sequencing of a conserved DNA locus in accordance with [ 11 , 25 ]. A proteomic analysis was conducted using matrix-assisted laser desorption ionization time-of-flight (MALDI-TOF) mass spectrometry with the BiotyperMicroflex LT instrument (Bruker Daltonics, Bremen, Germany).…”

“…The fermentation medium underwent filtration through filter papers, and the remaining feather residue underwent thorough washing with distilled water, followed by drying and subsequent weighing to calculate the extent of weight loss. The results are presented as a percentage relative to the initial dry weight of the feather [23,24].…”
