Two highly related crystal protein genes from Bacillus thuringiensis subsp. kurstaki HD-1, designated cryIIA and cryIIB (previously named cryBl and cryB2, respectively), were used to study host range specificity. Their respective gene products are 87% identical but exhibit different toxicity spectra; CryIIA is toxic to both mosquito and tobacco hornworm larvae, whereas CryIIB is toxic only to the latter. Hybrids of the crylIA and crylIB genes were generated, and their resultant gene products were assayed for toxicity. A short segment of CrylA-corresponding to residues 307 through 382 was shown to be sufficient for altering host range specificity-i.e., when this region replaced the corresponding segment of CryIIB, the resulting hybrid protein acquired toxicity against mosquitoes. The CryHA and CryHIB polypeptides differ by only 18 amino acds in this region, indicating that very few amino acid changes can have a substantial effect on the toxicity spectra of these proteins.

In this study, genomic DNA fi ngerprints of 40 Bacillus thuringiensis serovars representing different serotypes were obtained by Randomic Amplifi cation of Polymorphic DNA (RAPD-PCR) using three different randomic primers. The electrophoretic analysis of RAPD band profi les showed the presence of polymorphism among the serovars and serotypes. Between the primers used in this study, primer 2 was the most discriminatory and, therefore, was used to build the dendrogram. The similarity analysis of the electrophoretic profi les showed low level of similarity (16%) among the different B. thuringiensis serovars. The serovars were divided into two main groups, and these indicated a high level of genetic divergence both intra and inter-group. Results suggested that genomic fi ngerprints obtained by RAPD can be used for the genotypic characterization and identifi cation of B. thuringiensis serovars as a complement to fl agellar serology. ResumoO fi ngerprintings do DNA genômico baseado na amplifi cação aleatória de DNA polimórfi co (RAPD-PCR) de 40 sorovares de Bacillus thuringiensis representando diferentes sorotipos foi realizado usando três iniciadores randômicos. A análise eletroforética dos perfi s de RAPD revelou a incidência de polimorfi smo entre os sorovares e sorotipos. Entre os iniciadores utilizados, o iniciador nº 2 foi o mais discriminatório e, portanto, foi usado para construir um dendrograma. A análise da similaridade entre os perfi s eletroforéticos demonstrou baixo nível de afi nidade genética (16%) entre os diferentes sorovares de B. thuringiensis, agrupando esses sorovares em dois grupos principais com alta divergência genética intra e intergrupo. Logo, o RAPD baseado no fi ngerprintings genômico de B. thuringiensis pode ser usado na caracterização genotípica e identifi cação de sorovares de B. thuringiensis como um complemento à sorologia fl agelar.
