Isolation of a new set of Aspergillus nidulans mutants defective in nuclear migration

Abstract: In the filamentous fungus Aspergillus nidulans, nuclear migration in the germ tube is mediated by cytoplasmic dynein. We have previously reported the characterization of four nud (nuclear distribution) genes, nudA, nudC, nudF and nudG, involved in this process. The nudA and nudG genes respectively encode for the heavy chain and the 8-kDa light chain of cytoplasmic dynein. In this work, we describe an improved method for the isolation of nud mutants that has led to the identification of at least ten additional … Show more

“…In addition, nudG encodes a homologue of LC8 (ref. 23), which is one of the nud genes involved in nuclear migration in fungi 24,25 . Indeed, dynein, LIS1, NDEL1 and mammalian NUDC (mNUDC) are all nud genes 25 .…”

“…The Aspergillus orthologue of LIS1, NUDF, 145 functions within a complex of NUD proteins, including NUDA (dynein heavy chain), NUDG (dynein light chain), NUDK (dynactin subunit Arp1) and NUDE. [145][146][147][148][149] The composition of this complex is conserved in mammals, demonstrating that nuclear positioning is an indispensable function. Aspergillus NUDE has two mammalian orthologues termed NUDE and NUDE-like (NDE1 and NDEL1), which interact with LIS1.…”

“…Media, growth conditions, genetic crosses, A. nidulans transformations, and genomic DNA isolation were as described previously (Xiang et al 1995a,b;Willins et al 1995Willins et al , 1997. In many experiments described in this article, we transformed A. nidulans strains in which the A. nidulans Ku70 homolog was deleted (DnkuA) (Nayak et al 2006), with individual genomic fragments plus the plasmid pAid containing the pyrG selective marker (Xiang et al 1999). The DnkuA strains made most of the described experiments feasible due to an increased frequency of homologous integration of a genomic fragment into the A. nidulans genome (Nayak et al 2006).…”
