Isolation, characterization, and mesodermic differentiation of stem cells from adipose tissue of camel (Camelus dromedarius)

Abstract: Adipose-derived stem cells are an attractive alternative as a source of stem cells that can easily be extracted from adipose tissue. Isolation, characterization, and multi-lineage differentiation of adipose-derived stem cells have been described for human and a number of other species. Here we aimed to isolate and characterize camel adipose-derived stromal cell frequency and growth characteristics and assess their adipogenic, osteogenic, and chondrogenic differentiation potential. Samples were obtained from fi… Show more

“…MSC populations obtained from different tissues exhibit significant differences in their proliferation, differentiation, and molecular properties, which should be taken into consideration when planning their use in clinical protocols [55]. Further investigations for elucidation of molecular characteristics of skin stem cells are required with respect to adipose tissue stem cells [24] and cumulus cells [23].…”

“…Recently, the camel stem cell (SC) research has attracted some attention [23][24][25]. These studies isolated stem cells from cumulus cells [23], adipose tissue [24], and camel embryos [25]. Currently, the main targets of researchers' interest are epithelial and fibroblast differons of the skin.…”

“…T c was found after plotting the derived graph of heat flow to temperature diagram and finding heat flow peaks in this range. Approved experimental protocol was used to study the in vitro biocompatibility (Ref [21][22][23][24]. Cu 0.5 Ni 0.5 NPs were first sterilized in an autoclave and then dispersed in DulbeccoÕs modified EagleÕs medium (DMEM) using ultrasonic irradiation for 20 min.…”

“…Human adipose-derived mesenchymal stem cells (hAMSCs), rat adipose-derived mesenchymal stem cells (rAMSCs) and human USSCs were isolated as previously described. [25][26][27][28] For characterization of multipotent stem cells, after two passages, expression of surface markers was estimated by reaction with monoclonal antibodies (Ab) against rat and human CD34 (BD Pharmingen, San Diego, CA), CD45 (Abcam, Cambridge, UK), CD44 (BioLegend, San Diego, CA), CD90 (BD Pharmingen, San Diego, CA), CD105 (Abcam, Cambridge, UK) and human CD73 (BD Pharmingen, San Diego, CA).The cells were detached by trypsin/EDTA and incubated with the specific Ab and isotype controls in 100 mL 3% bovine serum albumin (BSA) in phosphate-buffered saline (PBS) for 1 h at 48 degrees C. About 10 5 -10 6 cells were used for each Ab. The cells were then fixed with 1% paraformaldehyde and analyzed with either an Epics XL (Beckman Coulter, Miami, FL) or a PAS (Partec M€ unster, Germany) machine.…”
