Isolation, Characterization and investing the Industrial Applications of Thermostable and Solvent Tolerant Serine Protease from Hot Spring Isolated Thermophililic Bacillus licheniformis U1

Abstract: Protease is the largest selling enzyme in the world due to its various applications in the making of detergent, food and leather, meat tenderisation and pharmaceutical industries. The aim of the study is to isolate and identify thermophilic Bacillus licheniformis U1 strains for thermostable protease production. The partial purified enzyme was characterized under different conditions using Anson-Hagihara’s method. Casein as a substrate in the concentration of 0.6 % w/v optimum for enzyme activity and tolerant u… Show more

“…Spore-forming-rod-shaped, the Gram-positive bacterium was isolated from the Unnai hot spring (N 20° 51’ 14.1091" and E 73° 20’ 9.4986"), Gujarat, India. The isolate previously identified as a Bacillus licheniformis U1 [ 48 ]. The hot spring is natural thermal habitat, that harbour diverse thermophilic bacteria and a potential source of thermostable laccase [ 54 , 55 ].…”

“…The strain was identified based on partial gene sequence of 16S rRNA, and the sequence submitted to GenBank (Accession No. GU979026) [ 48 ]. The isolate maintained on nutrient agar, pH 8.0 and subculture after every 30 days on same solid media.…”

“…The strain Bacillus cohnii was isolated from the less explored Unnai hot spring, where the temperature usually reported between 57°C to 61°C. Earlier, thermophilic protease producing Bacillus licheniformis was reported from the Unnai hot spring (Dudhagara et al, 2014). Bacillus species are attractive industrial tools for a variety of reasons, including their high growth rates leading to short fermentation cycle times, their capacity to secrete proteins into the extracellular media and the Generally Regarded As Safe (GRAS) status with the food and drug administration (Manikandan et al, 2014).…”

“…Hot springs are the rich source for the thermophilic bacteria for the direct isolation of thermostable enzymes (Olajuyigbe and Kolawole, 2011). Recently, fewhot spring isolated thermophilic Bacillus species are well studied for the alkaline protease production including, Bacillus licheniformis (Dudhagara et al, 2014), Bacillus subtilis (Priya and Archana, 2008) and Geobacillus sp. (Wei et al, 2007).…”
