Isolation, characterization and evaluation of 11 autosomal STRs suitable for population studies in black and gold howler monkeysAlouatta caraya

Abstract: We identified 11 polymorphic microsatellite markers for Alouatta caraya . Three markers were isolated from an enriched genomic library of A. caraya (AC14, AC17, AC45), five were previously described in Homo sapiens (TGMS1, TGMS2, D5S117, D8S165, D17S804), and three were identified for Lagothix lagotricha (1110, 1118, 157). Forty-eight individuals from one Argentinean population were genotyped, yielding heterozygosity values between 0.146 and 0.792. These markers provide an exclusion power of 0.922 when neither… Show more

“…On the other hand, comparative studies of the population from 1982 to 2004 suggest that howlers can cope relatively well with moderate deforestation and fragmentation of the forest [5,15]. However, recent studies indicate an incipient process of isolation for the population of howlers living in an area of 5,000 ha surrounding the EBCo [17]. Based on genetic data, Oklander [16] suggests that habitat fragmentation may serve to decrease the ability of howlers to disperse between groups.…”

“…In addition to this, the EBCo academic staff develops studies on the The forests surrounding the EBCo are selectively logged and clear cut, and the landscapes are characterized by isolated forest patches of 10-15 ha located in a matrix of grasslands. The average isolating distance between forest patches is 1,763.2 ± 538.7 m (N = 11 fragments) [16,17].…”

“…For each sample, we amplified 11 microsatellites characterized for A. caraya: AC14, AC17, AC45, TGMS1, TGMS2, D5S117, D8S165, D17S804, LL1110, LL1118, LL157 [Di Fiore & Fleischer, 2004;Tomer et al, 2002]. These microsatellites were already screened for this population in Oklander et al [2007]. All loci were in Hardy-Weinberg equilibrium and mean observed heterozygosity was 0.54 (range 0.146-0.792) .…”

“…All PCR reactions were performed in a GeneAmp® PCR System 9700 (Applied Biosystems). PCR primers´ sequences are listed in Table 3.3.4.1 and have been designed with M13 tails for product detection with M13 primers labeled with FAM™, as previously described (Oklander, Zunino et al 2007). After PCR, products were prepared with 7.5µL Hi-Di Formamide, 1µL…”
