Isolation and screening of cellulolytic fungi from Salacca zalacca leaf litter

Abstract: Cellulases are the main enzymes in a bioconversion of lignocellulosic biomass. Fungal cellulases have been proven to be better candidates for this process than other microbial cellulases since they secrete free cellulase complex including endoglucanases, cellobiohydrolases, and β-glucosidase. This study was conducted to isolate and screen of cellulolytic fungi from Salacca leaf litter. There were about 12 fungal isolates which were obtained in this research and 8 isolates showed cellulolytic activity. Based on… Show more

“…There were two Aspergillus isolates from rhizosphere of raru songal, two isolates from rhizosphere of raru dahanon and one isolate from rhizosphere of raru pulut, while for Penicillium, there were two isolates from rhizosphere of raru songal and each one isolated from rhizosphere of raru dahanon and raru pulut. Aspergillus and Penicillium are cellulolytic fungi which are mostly isolated by previous researchers both from the soil and from leaf litter (Herculano et al 2011;Reddy et al 2014;Torres et al 2014;Sari et al 2017). Molecular identification revealed five Aspergillus isolates consisted of three species, namely, Aspergillus sp.…”

“…Fungi banyak digunakan pada produksi enzim karena memiliki harga yang rendah dengan produktivitas tinggi, tingkat produksi yang cepat dan kemungkinan modifikasi enzim yang mudah. Selulase biasa digunakan dalam produksi tekstil, kertas dan tisu, laundry dan detergent, pertanian, pengobatan dan industri makanan (Jayasekara & Ratnayake, 2019;Sari et al, 2017).…”

“…The screening was done by inoculating the fungal disk onto the SMA plate and incubated at 27ºC for 96 h. The hydrolysis zone around the colony indicates protease activity due to the casein hydrolysis process (Kamath et al 2010;Maitig et al 2018). The enzymatic activity was determined by clear zone formation around the fungal disk (Lusi et al 2017). The enzymatic index (EI) was measured as a semi-quantitative estimate of the enzyme activities, according to the formula below (Coronado-Ruiz et al 2018;Maitig et al 2018).…”
