Isolation and Properties of Flagella of Trypanosomatids*

Abstract: A procedufe is described for the isolation of flagella of Crithidia fasciculata, Herpetomonas samuelpessoai and Leishmania tarentolae in a highly purified state and giving reasonably good yield. The 3 types of flagella give a similar electrophoretic pattern of proteins. It is shown that H. samuelpessoai and, to a lesser extent, C. fasciculata flagella confer protection against Trypanosoma cruzi infection.

“…Figure 1 represents a summary of the fractionation scheme. Many of the classic de¯agellation and deciliation methods previously used by other investigators for eukaryotic systems (Adoutte et al 1980;Dentler 1980;Gualtieri et al 1986;Hyams 1992;Pereira et al 1977;Piperno and Luck 1976;Rosebaum and Child 1967;Satir et al 1976;Witman 1986;Witman et al 1972) were initially employed, but none worked for Trichomonas vaginalis with the exception of mechanical shearing (Rosebaum and Child 1967). Flagella from the pH-sucrose method (Rosebaum and Child 1967) became swollen with vigor.…”

“…Of these, at least the first two can change in relative quantity during cytodifferentiation and hence the tubulin biosynthesis. Previous identification of tubulin has been based on less stringent evidence-e.g., electrophoretic mobility of protein bands from L. tarentolae flagella (37), from L. donovani surface membranes (38), and comigration with rat brain tubulin for Trypanosoma cruzi flagella (39). Tubulin biosynthesis occurs rapidly in amastigote-to-promastigote transformation long before cytological changes become noticeable.…”

“…The FLAG1 protein was purified from Leishmania flagella preparations using previously described methods (Pereira et al 1977, Ismach et al 1989). The flagellar-enriched fraction was extracted using Brij-97, and nonsoluble material was eliminated by centrifugation at 39,000 · g for 1 h at 4°C.…”
