Isolation and Molecular Characterization of the Fire Blight Pathogen, Erwinia amylovora, Isolated from Apple and Pear Orchards in Egypt

Abstract: Eleven Erwinia amylovora isolates, the causal agent of the fire blight disease, were isolated from apple orchards adjacent to those of pears showing fire blight disease symptoms. Initial characterization concerning morphological and biochemical traits revealed that all isolates were typical to those of E. amylovora. Furthermore, all bacterial isolates showed typical symptoms of the fire blight disease upon infection of unripe pear fruits. Molecular characterization of these isolates via PCR utilizing specific … Show more

“…Genomic DNA was extracted using CTAB/NaCl method ( Ashmawy et al, 2015 ). The bacterial isolates were identified by PCR of the 16S rDNA gene sequence using fD2 (5′- AGAGTTTGA TCCTGGCTCAG-3′) and rP1 (5’-ACGGTTACCTTGT TACGACTT-3′) primers ( Weisburg et al, 1991 ).…”

“…Ten most virulent and important bacterial plant pathogens based on their pathogenicity level, economic impact, and molecular aspects were listed1. Most devastating bacterial plant pathogens species were isolated in Egypt which belong to Pseudomonas syringae pathovars 3,4,5,6 , Ralstonia solanacearum 7,8,9 , Agrobacterium tumefaciens 9,10,11,12 , Xanthomonas 3,13,14 , Erwinia amylovora 15,16,17,18,19,20 and Xylella fastidiosa 21. Bacterial soft rot soft rots commonly affect vegetables and fruits. It can occur on crops in the eld and on the market.…”
