Metronomic chemotherapy refers to the close, regular administration of comparatively low doses of cytotoxic drugs, with minimal or no drug-free breaks, over prolonged periods. It is thought to have an antiangiogenic basis. However, whereas surprisingly durable and potent tumor responses have been observed in a number of preclinical tumor models, relapses usually eventually occur using this type of treatment strategy. We therefore decided to test modified metronomic chemotherapy regimens that might significantly delay such relapses, but still maintain modest and acceptable toxicity profiles. Here, we show that repeated administration of bolus doses (BDs) of cyclophosphamide every 3 or 6 weeks, combined with a daily oral low-dose metronomic (LDM) regimen (20 mg/kg/d cyclophosphamide), improves efficacy and significantly delays progression of transplanted PC-3 human prostate cancer xenografts, syngeneic transplanted EMT-6 breast tumors, and ''spontaneous'' murine erythroleukemia. Efficacy was superior whereas toxicity was mild and comparable to the LDM regimen, the latter assessed by body weight, neutrophil, lymphocyte, and total white blood counts. Antiangiogenic activity, measured by reduction in circulating endothelial precursor cells, revealed that the greatest degree of suppression occurred using the combination treatment. Overall, our results indicate that the administration of intermittent BD combined with chronic oral LDM cyclophosphamide is a potent treatment regimen for controlling tumor growth, which has a low toxicity profile, over prolonged periods of time. The retroviral integration siteThe hematopoietic system is composed of a hierarchy of cells, ranging from the pluripotent stem cell, committed myeloid and lymphoid progenitor cells, to terminally differentiated blood cells (Metcalf 1988;Sachs 1987). The molecular mechanisms that control the developmental and proliferative decisions of the pluripotent hematopoietic stem cell are largely unknown. The extreme cellular heterogeneity of hematopoietic populations, combined with the low frequency of stem cells, has hampered experimental approaches to this problem. The generation of mutations that affect stem cell function and the molecular analysis of existing developmental mutations that control hematopoiesis (Pawson and Bernstein 1990) provide two powerful genetic strategies for identifying and characterizing the genes that control normal blood cell formation. In addition, because cellular genes involved in leukemic transformation may also have a role in regulating normal stem cell function, the identification of such genes may provide insights into both normal and leukemic hematopoiesis. Transformation by certain retroviruses can involve the integration of proviral DNA near or within specific cellular genes. Such integration events most frequently lead to the elevated expression of these genes (for review, see in Peters 1990), although the tumor suppressor gene p53 can also be inactivated by retroviral integration of the Friend leukemia virus (Ben-David et al. 1988, 1990b. The analysis of common retroviral integration events can, therefore, be used to identify and clone novel transforming genes involved in tumorigenesis. Using this approach, a large number of genes important in the leukemic transformation of hematopoietic cells, including have been isolated (Peters 1990).The Fli-1 and Spi-1 genes have been shown by us and others (Moreau-Gachelin et al. 1988; Ben-David et al. 1990) to be involved in erythroleukemia induction by various strains of Friend leukemia virus. Bi is identical to the proto-oncogene Spi-1IPU.1 (Spi-l), an ets family member. Protein-DNA contacts are shown to resemble those of the helix-turn-helix homeodomain proteins. By Northern (RNA) analysis, we found that Spi-1 mRNA is present at low levels during murine CFU-E maturation and is at least 20-fold higher in uninduced MEL, a transformed proerythroblast-like cell line which contains an activating/transforming insertion of spleen focus-forming virus at the Spi-I locus. Dimethyl sulfoxide-induced MEL cell differentiation decreases Spi-) mRNA to approximately 20%o of the uninduced level before commitment occurs. In addition to erythroid cells, Spi-I mRNA is present in B cells, myelomonocytes, and mast cells but not in T cells and nonhematopoietic cell types. In situ hybridization demonstrated Spi-l mRNA expression in bone marrow, spleen, interstitial nonhepatocytes of the liver, and interstitial nontubular cells of the testis. The Spi-) locus was mapped on human chromosome 11 to the same interval as ACP2 (lysosomal acid phosphatase), between the anonymous DNA markers D11S33 and D11S14. This region has not yet been found to be associated with a human malignancy.We have previously identified by electrophoretic mobility shift analysis (EMSA) several factors in murine erythroleukemia (MEL) cells that bind to specific sequences within the mouse n-major (3M)-globin intervening sequence 2 (IVS2) (27). The region encompassing these sites has been observed by others to be a tissue-specific DNase I-hypersensitive site and the border of nucleosome phasing on the globin gene in erythroid cells (7,39,78). This region includes two homologous sites for factor B1 (B1-A and B1-B) and one site each for factors Oct-1 (40) and B2 (now designated We report here the cloning of the cDNA for MEL factor B1 by using a modification of the approach taken by Tsai et al. (83), involving a mammalian expression system combined with EMSA analysis of extracts from the transfected COS cells as the detection method. The cDNA sequence revealed that factor B1 is identical to the proto-oncogene/DNAbinding factor Spi-1/Pu.1ISfpi-1 (Spi-1 stands for spleen focus-forming virus [SFFV] proviral integration 1) (33,47,55,65), an ets gene family member which had been previously cloned by different methodologies.
