Isolation and Immunochemical Localization of Collagenase in Mouse Peritoneal Macrophages1

Abstract: Collagenase was isolated from the culture medium of thioglycollate-stimulated mouse peritoneal exudate macrophages. The macrophage collagenase activity was inhibited by goat anti-mouse bone collagenase antibody, indicating that macrophage collagenase immunologically cross-reacts with mouse bone collagenase. The enzyme was localized in mouse peritoneal macrophages by indirect immunofluorescent antibody technique. Distinct granular fluorescence was observed intracellularly in most thioglycollate-stimulated macro… Show more

“…Sakamoto has described the preparation of a goat antiserum against mouse bone collagenase, that reacts strongly with peritoneal macrophages (42). Using the avidin-biotinperoxidase complex (ABC) method, the same antibody showed cross-reactivity with purified collagenase obtained from culture media of rat tumor epithelial cells, suggesting that mouse bone collagenase and rat mammary tumor collagenase were antigenically closely related.…”

“…Although collagenase is produced by a variety of mammalian cell types (6,7), the mechanisms governing collagenase synthesis and secretion differ. In polymorphonuclear leukocytes (8,9) and monocytes/macrophages (10)(11)(12), collagenase is synthesized and stored within the cell in granules; secretion is regulated by factors controlling granule release rather than by those controlling enzyme synthesis (7). In contrast, fibroblast (e.g., skin and synovial cell) collagenase is not stored.…”
