Pueraria tuberosa (Roxb. ex Willd.) DC., known as Indian Kudzu belongs to family Fabaceae and it is solicited as “Rasayana” drugs in Ayurveda. In the present study, we analyzed the efficacy of an ethyl acetate fraction from the tuber extract of Pueraria tuberosa (fraction rich in antioxidant compounds, FRAC) against menopausal osteoporosis, and breast and ovarian cancer cells. The FRAC from Pueraria tuberosa was characterized for its phenolic composition (total phenolic and flavonoid amount). Antioxidant property (in vitro assays) of the FRAC was also carried out followed by the analysis of the FRAC for its antiosteoporotic and anticancer potentials. The antiosteoporotic activity of FRAC was investigated in ovariectomy-induced osteoporosis in rats. The cytotoxicity effect was determined in breast and ovarian cancer cells. Gas chromatography/mass spectrometry (GC/MS) analysis of the FRAC was performed to determine its various phytoconstituents. Docking analysis was performed to verify the interaction of bioactive molecules with estrogen receptors (ERs). The FRAC significantly improved various biomechanical and biochemical parameters in a dose-dependent manner in the ovariectomized rats. FRAC also controlled the increased body weight and decreased uterus weight following ovariectomy in rats. Histopathology of the femur demonstrated the restoration of typical bone structure and trabecular width in ovariectomized animals after treatment with FRAC and raloxifene. The FRAC also exhibited in vitro cytotoxicity in the breast (MCF-7 and MDA-MB-231) and ovarian (SKOV-3) cancer cells. Furthermore, genistein and daidzein exhibited a high affinity towards both estrogen receptors (α and β) in the docking study revealing the probable mechanism of the antiosteoporotic activity. GC/MS analysis confirmed the presence of other bioactive molecules such as stigmasterol, β-sitosterol, and stigmasta-3,5-dien-7-one. The FRAC from Pueraria tuberosa has potential for treatment of menopausal osteoporosis. Also, the FRAC possesses anticancer activity.

Menopause didefinisikan sebagai 1 tahun tanpa menstruasi. Wanita menopause kemungkinan berisiko mengalami Nonalcoholic Fatty Liver Disease(NAFLD). Efek menguntungkan dari antosianin telah dibuktikan dalam penelitian hewan,sehubungan dengan stres oksidatif dan steatosis hati. Ubi jalar ungu mengandung antosianin yang berfungsi sebagai antioksidan. Penelitian ini dilakukan dengan menggunakan sampel tikus yang sudah di ovariektomi, kemudian diberikan ekstrak etanol ubi jalar ungu pada tikus secara oral selama 30 hari. Pada hari pertama dilakukan randomisasi dibagi menjadi empat kelompok yaitu kelompok kontrol negatif (P0), kelompok perlakuan pemberian ekstrak etanol ubi jalar ungu dosis 1ml (P1), dosis 2 ml (P2), dan dosis 4ml (P3). Pada hari ke-31 dilakukan terminasi untuk di ambil jaringan hati dan pembuatan preparat histologi, kemudian dilakukan pengamatan pada preparat dengan pembesaran 10x10 pada 2 lapang pandang. Dari 28 sampel hati tikus yang diperiksa, didapatkan semua sel hati tikus mengalami degenerasi lemak atau steatosis sebanyak >5% yang merupakan kunci dari penyakit hati berlemak nonalkohol (NAFLD), namun jumlahnya berbeda-beda. Rerata perlemakan kelompok kontrol negatif didapatkan hasil perlemakan paling tinggi yaitu 49,1%, kelompok perlakuan pemberian dosis 1ml, 2ml, dan 4ml didapatkan hasil 39,48%, 31,62%, dan 29,14%. Setelah diuji secara statistik didapatkan perbedaan yang signifikan (p<0,05) terhadap pengaruh pemberian ekstrak etanol ubi jalar ungu.Berdasarkan hasil penelitian yang telah dilakukan, maka dapat disimpulkan bahwa pemberianekstrak etanol ubi jalar ungu selama 30 hari dapat menurunkan degenerasi lemak pada hati tikus yang mengalami penyakit hati berlemak nonalkohol (NAFLD). Pediatric solid tumors (PSTs) are life-threatening and can lead to high morbidity and mortality rates in children. Developing novel remedies to treat these tumors, such as glioblastoma multiforme and sarcomas, such as osteosarcoma, and rhabdomyosarcoma, is challenging, despite immense attempts with chemotherapeutic or radiotherapeutic interventions. Soy (Glycine max) and kudzu roots (KR) (Pueraria spp.) are well-known phytoestrogenic botanical sources that contain high amounts of naturally occurring isoflavones. In the present study, we investigated the antioxidant and cytotoxic effects of the extracts of KR and soy molasses (SM) against PSTs. The green extraction of isoflavones from KR and SM was performed using natural deep eutectic solvents. The extracts were subsequently analyzed by high-performance liquid chromatography (HPLC)-diode array detector (DAD) coupled with high-resolution (HR) mass spectrometry (MS), which identified 10 isoflavones in KR extracts and 3 isoflavones in the SM extracts. Antioxidant and cytotoxic activities of KR and SM extracts were assessed against glioblastoma multiforme (A-172), osteosarcoma (HOS), and rhabdomyosarcoma (Rd) cancer cell lines. The KR and SM extracts showed satisfactory cytotoxic effects (IC50) against the cancer cell lines tested, particularly against Rd cancer cell lines, in a dose-dependent manner. Antioxidant activity was found to be significantly (p ≤ 0.05) higher in KR than in SM, which was consistent with the results of the cytotoxic activity observed with KR and SM extracts against glioblastoma and osteosarcoma cells. The total flavonoid content and antioxidant activities of the extracts were remarkably attributed to the isoflavone content in the KR and SM extracts. This study provides experimental evidence that HPLC-ESI-HRMS is a suitable analytical approach to identify isoflavones that exhibit potent antioxidant and anticancer potential against tumor cells, and that KR and SM, containing many isoflavones, can be a potential alternative for health care in the food and pharmaceutical industries.
