Identification of 33 Bacillus spp. isolates from different environmental samples collected from the territory of Serbia was performed by sequencing of the 5ʹ-hypervariable section of 16S rRNA gene. Eight species were identified within four phylogenetic groups: B. pumilus, B. megaterium, B. subtilis and B. cereus. Determination of their antibiotic resistance was performed using the minimum inhibitory concentration (MIC) assay. We found that just one isolate was resistant to gentamicin, 9 were resistant to clindamycin and all were resistant to vancomycin. Based on the profile of resistance, the isolates were categorized into 4 categories. In silico analysis of the erythromycin-resistance (erm) gene for clindamycin resistance showed their distribution between related and nonrelated soil and human isolates including different species of Bacillus genera. This finding indicates that Bacillus spp. from the environment could be a source of resistance to clindamycin. The potential for the presence and spread of resistance determinants in the soil and similar ecosystems exists so that monitoring of antibiotic resistance genes in nonpathogenic Bacillus strains from the environment is advised.

In the global frame aiming at assessing bacterial susceptibility for safer and cost-effective healthcare, the present survey was conducted in three hospitals: Bafoussam Regional Hospital (BRH), Bangwa Protestant Hospital (BPH) and Bangangté District Hospital (BDH). Sampling was performed by fingerprinting on culture media and swabbing of hospital devices or surfaces. Wards of interest included: Pediatrics, Medicine, Operating Theater, Intensive Care Unit, Maternity, and, in the BDH, Laboratory in addition. Culture, isolation, identification and susceptibility tests were conducted according to standard guidelines and assigned contamination rates. Seventeen antibacterial agents were chosen and included representatives of major families of antibiotics used in Cameroon. Analysis of 238 specimens revealed 90%, 86% and 92% contamination rates in the BRH, BPH, and BDH, respectively. On healthcare provider's hands, the respective rates were 63%, 100% and 91%. Bacillus and Staphylococcus were predominant bacteria types in all settings (BPH: 92%; BDH: 86%; BRH: 81%). Susceptibility profiles indicated high resistance rates and clonal distribution in all settings; and most reduced susceptibility with common drugs. Further investigations and previous works alleged drug use and basic hygiene as crucial in addressing resistance issues for safer care. This would be achieved with State support to public and private institutions.

Background: Candida infection currently presents a major problem. The natural products from endophytic bacteria are thought to have a strong potential for treatment. Objectives: The present study aimed to isolate endophytic bacteria from Dryopteris uniformis, its identification, and investigation of their anticandidal activity and confirm it. Methods: The isolated endophytic bacteria from D. uniformis were identified by using 16S rRNA gene sequencing. The anticandidal assay of endophytic bacteria was performed by using the patch method. Endophytic bacteria were successively fractionated by using different polarity solvents. The anticandidal activity of each solvent fraction was evaluated by using the disc diffusion method and was confirmed by scanning electron microscopy. Results: Fifty-one endophytic bacteria were isolated from the lead and stem/root of D. uniformis and screened for anticandidal activity against five Candida species, Candida saitoana (KACC 41238), C. albicans (KACC 30062), C. albicans (KACC 30003), C. glabrata (KBNO6P00368) and C. glochares (KACC 30061). Among these, six endophytic bacteria exhibited strong anticandidal activity with an inhibition zone diameter between 9.29 and 47.67 mm against four Candida species; these bacteria were identified as Burkholderia sp. UR 1-07 (DUS14), Staphylococcus sp. WW60 (DUS56), Bacillus sp. cryopeg (DUS59), Paenibacillus sp. rif200865 (DUL128), Staphylococcus warneri (DUS130), and Bacillus psychrodurans (DUS131) using 16S rRNA gene sequencing. The minimum inhibitory concentration (MIC) of the butanol fraction of the selected endophytic bacteria, Burkholderia sp., was 250 µg/mL and the minimum fungicidal concentration (MFC) was 500 µg/mL. Scanning electron microscopy result indicated that, MIC of promising endophytic bacteria Burkholderia sp. destroyed the target Candida cells. Conclusions: This study demonstrated anticandidal potential of endophytic bacteria derived from D. uniformis.
