Isolation and Genotyping of Acanthamoeba Strains from Soil Sources from Jamaica, West Indies

Abstract: Acanthamoeba spp. are opportunistic pathogens that are ubiquitous in nature. Many species of this genus are responsible for a fatal encephalitis and keratitis in humans and other animals. Seventy-two soil samples were collected from the parishes across Jamaica and assessed for the presence of Acanthamoeba spp. Cultivation was carried out on non-nutrient agar plates seeded with heat killed Escherichia coli. PCR and sequencing of the DF3 region were carried out in order to genotype the isolated strains of Acanth… Show more

“…Acanthamoeba spp. are the most common free-living protozoa, isolated from public water supplies, swimming pools, bottled water, distilled water bottles, ventilation ducts, the water-air interface, air-conditioning units, soil, beaches, vegetables, air, surgical instruments, and contact lenses, indicating the ubiquitous nature of these organisms (Cruz and Rivera, 2014;Todd et al, 2015;Al-Herrawy et al, 2015). Acanthamoeba species can cause various diseases such as keratitis and granulomatous amoebic encephalitis, which is often fatal in immune-compromised as well as immunocompetent patients (Król-Turmińska and Olender, 2017).…”

“…The strains were isolated from the soil collected near electroplating plants from Shandong, Jiangxi, Jilin, Liaoning, and Heilongjiang provinces. Briefly, 1 g of soil of each sample was dissolved in 10 mL sterile water, and then 100 µL of this soil suspension was added to LB medium with 2 mM Cr(VI) [15,35]. After incubating for 72 h at 37 • C, with agitation at 180 rpm, the enriched culture solution was then spread-plated on LB-solid medium with 2 mM Cr(VI) [35].…”
