Access to safe water is essential to life and public health of human. Nearly, 884 million people about one-sixth of the world's populations are consuming contaminated water. In this study, the occurrence and the molecular characterization of enteric viruses particularly; enteric adenovirus, rotavirus and free-living protozoan Acanthamoeba spp. were determined in 180 tap water samples collected from five Egyptian governorates during December 2016 to November 2017. Moreover, we aimed to verify whether these isolated Acanthamoeba spp. could play a role in transmission of enteric adenovirus and rotavirus. Multiplex semi-nested RT-PCR, PCR and sequencing were used to detect and genotype rotavirus, enteric adenovirus and Acanthamoeba. Based on the sequence analysis of the Diagnostic Fragment 3 (DF3) of small subunit rRNA gene, Acanthamoeba isolates were genotyped. The present study showed that 26.7% (48/180) of tap water samples were positive for Acanthamoeba spp. The highest occurrence rate of Acanthamoeba (36.1%) was detected in tap water samples collected from Faiyum governorate. On the other hand, viral genomes were detected in 44 samples. Rotavirus and enteric adenovirus were observed in 15.6% (28/180) and 8.9% (16/180) of samples, respectively. Qalyubia and Faiyum governorates showed high level of viral contamination up to 50% (18/36) and 47.2% (17/36), respectively. Winter was the peak season for viral and Acanthamoeba contamination in tap water. Mixed contamination between at least one virus and Acanthamoeba was observed. Acanthamoeba lenticulata, Acanthamoeba T4, rotavirus G1P [8], and human enteric adenovirus 41 were the most predominant types identified during this work. The prevalence of pathogenic Acanthamoeba genotypes, especially genotype T4 and enteric viruses (rotavirus and adenovirus) in the examined tap water samples presented health hazards to consumers.

Chromium contamination has been an increasing threat to the environment and to human health. Cr(VI) and Cr(III) are the most common states of chromium. However, compared with Cr(III), Cr(VI) is more toxic and more easily absorbed, therefore, it is more harmful to human beings. Thus, the conversion of toxic Cr(VI) into Cr(III) is an accepted strategy for chromium detoxification. Here, we isolated two Bacillus cereus strains with a high chromium tolerance and reduction ability, named B. cereus D and 332, respectively. Both strains demonstrated a strong pH and temperature adaptability and survival under 8 mM Cr(VI). B. cereus D achieved 87.8% Cr(VI) removal in 24 h with an initial 2 mM Cr(VI). Cu(II) was found to increase the removal rate of Cr(VI) significantly. With the addition of 0.4 mM Cu(II), 99.9% of Cr(VI) in the culture was removed by B. cereus 332 in 24 h. This is the highest removal efficiency in the literature that we have seen to date. The immobilization experiments found that sodium alginate with diatomite was the better method for immobilization and B. cereus 332 was more efficient in immobilized cells. Our research provided valuable information and new, highly effective strains for the bioremediation of chromium pollution.

Morphological and molecular identification of free living amoeba isolated from hospital water in Tunisia

Free-living amoebae (FLA) are opportunistic and ubiquitous protozoa that are widely found in various environmental sources. They are known to cause serious human infections. The aim of our study was to detect FLA and Acanthamoeba spp. in hospital water circuits. Eighty-four water samples were collected over a period of 4 months (September-December 2011) from different wards of the Sfax University Hospital (surgical services, intensive care unit, operating theater, and water storage tanks). FLA were detected in 53.5 % of samples as follows: surgical services (80 %), operating theater and surgical intensive care unit (13.3 %), medical intensive care unit (0 %), water storage tanks (6.6 %). The predominant morphotype was the acanthopodial (89 %). The others morphotypes were as follows: monopodial (40 %), dactylopodial (22 %), rugosa (62 %), eruptive (24 %), fan shaped (18 %), and polypodial (18 %). Acanthamoeba was found in 40 samples (47.6 %). 64.2 % of isolates were identified as Acanthamoeba spp. by PCR, using primers to amplify a region of 18S rDNA which showed variation in the product length. Sequence analysis of five PCR products identified Acanthamoeba sp. These isolates belong to T4, T10, and T11 genotypes, and to our knowledge this is the first report of the T10 and T11 genotype in Tunisia.The occurrence of potentially pathogenic FLA in the hospital environment may represent a health risk for patients, since these organisms can cause severe opportunistic illness and also can harbor pathogenic agents. Thus, increased awareness regarding these parasites and recognition of their importance, particularly in immunocompromised patients is crucial.
