Isolation and culture of protoplasts from young sporophytes ofSalvinia natans aseptically obtained by co-culture of female and male gametophytes

Abstract: Sporophytes were aseptically obtained by co-culture of female and male gametophytes derived from two types of spores (megaspores and microspores) of the heterosporous fern Salvinia natans All. Protoplasts isolated enzymatically from juvenile leaflets of sporophytes were cultured in a 1/10 Murashige and Skoog's medium containing 2.2 ktM naphthalene acetic acid, 2.2 ~tM 6-benzyl-aminopurine, 0.35 M mannitol, and 0.05 M sucrose. Cell division took place within 6 days of culture, and cell-clusters composed of 9-10… Show more

“…The number of shoots was reported stagnant on MS medium supplemented with Kin (0.5 mg/L), even after four months of incubation. Generally, ferns micropropagated using spores in most of the studies like Salvinia natans [32], Dryopteris affinis [33], Equisetum arvense [34], Marsilea minuta [35], and Pteris tripartita [36], but the shoots were induced from the nodal part of the rhizomes in present study. It has been reported that the supplementation of abscisic acid in the medium induced shoots, roots, and elongation of petioles of M. quadrifolia [37,38], whereas the supplementation of auxins has no ameliorative effects on the regenerative response in this plant species [28].…”

“…Enzyme treatment in particular has been used to generate protoplasts from the female gametophytes of a wide range of plant species including Ginkgo biloba, Salvinia natans, Laminaria digitata, Petunia hybrida (Laurain et al 1993, Nakamura and Maeda 1994, Benet et al 1997, Sangthong et al 2009, and Torenia fournieri (Mol 1986, Chen et al 2008.…”
