Isolation and Culture of Neonatal Mouse Cardiomyocytes

Abstract: Cultured neonatal cardiomyocytes have long been used to study myofibrillogenesis and myofibrillar functions. Cultured cardiomyocytes allow for easy investigation and manipulation of biochemical pathways, and their effect on the biomechanical properties of spontaneously beating cardiomyocytes. The following 2-day protocol describes the isolation and culture of neonatal mouse cardiomyocytes. We show how to easily dissect hearts from neonates, dissociate the cardiac tissue and enrich cardiomyocytes from the cardia… Show more

“…Although the use of animal models to study cardiac diseases has become standard in cardiovascular research [49], it is difficult to analyse Ca 2+ regulation in the whole hearts or animals. Recently, multiparametric electrophysiological imaging of the mammalian heart was developed to detect Ca 2+ transients in vivo; however, this measurement is difficult to apply under IH.…”

“…Primary cultures of neonatal mouse cardiomyocytes were generated as described previously [19]. Briefly, hearts collected from 1–2-day-old mice were washed, minced into small pieces(approximately 0.5– 1 mm 3 ) and predigested in isolation medium (0.5 mg/ml trypsin-HBSS solution) under gentle agitation overnight at 4°C.…”

“…NCMs were isolated from the hearts of 0-to 3-dayold neonatal mice as described previously (57). Briefly, ventricles from neonates were dissected, followed by 1.5 mg/ml Collagenase/Dispase (Roche Diagnostics) digestion.…”
