Isolation and characterization of turkey bone marrow‐derived mesenchymal stem cells

Abstract: Flexor tendon injury is often associated with suboptimal outcomes and results in substantial digit dysfunction. Stem cells have been isolated from several experimental animals for the growing interest and needs of utilizing cell‐based therapies. Recently, turkey has been developed as a new large animal model for flexor tendon research. In the present study, we reported the isolation and characterization of bone marrow‐derived mesenchymal stem cells (BMSCs) from 8‐ to 12‐month‐old heritage‐breed turkeys. The is… Show more

“…Far less is known about MSCs isolated from birds. Only a few groups have reported on the isolation of MSCs from turkeys and ducks ( 41 43 ), and although more research has been done studying the characteristics of MSCs derived from chickens ( 44 ), the effects of chicken MSCs, in particular the chicken MSC secretome, on wound healing has not been explored.…”

“…It is possible that RCECs in the young group show a strong phenotype of tendon/tendon stem cells already. In previous studies, Tnc was used as a marker for tendon differentiation, 38,39 but Tnc is not solely expressed in the tendon, being highly expressed in the bone and cartilage as well. 40,41 Our results show that Tnc was not upregulated after BMP12 induction.…”

“…Total RNA (1 µg/sample) was reverse transcribed (5 min at 20˚C, 20 min at 46˚C, 1 min at 95˚C and held at 4˚C) using a Reverse Transcription kit (Takara Bio, Inc.). qPCR was performed with the SYBR Green PCR Master Mix (Applied Biosystems; Thermo Fisher Scientific, Inc.) on a CFX96 Touch™ Deep Well Real-Time PCR Detection system (Bio-Rad Laboratories, Inc.) according to previous publications (47,48). Briefly, the thermal program consisted of 2 min at 95˚C, then 40 cycles of amplifications, 5 sec at 95˚C for denaturation, 5 sec at 65˚C for annealing, and 5 sec at 95˚C for extension.…”
