Isolation and Characterization of the Major Antigen of Rat Kidney

Abstract: The major antigen of rat kidney (KMA) was isolated by chromatography on DEAE-cellulose and Sephadex G-25 gel filtration. The sedimentation coefficient of the purified antigen was about 1.2 S and its electrophoretic mobility corresponded to that of α2-globulins. Its diffusion coefficient was estimated by the method of Allison and Humphrey as 11.4 × 10−7 cm2 sec−1. On gel filtration through Sephadex G-100, the purified preparation was resolved into two components with identical antigenic properties but of differ… Show more

“…Concerning more specifically the kidney, we have used in our laboratories immunochemical methods to identify several mouse and rat tissue antigens [19,20]: data have also been obtained by other workers in the calf [21] and in man [22,23]. Moreover, we have isolated and characterized the kidney major antigen (KMA) [24] and showed its nephritogenic effects in homologous rat [25]: E d g in g t o n et al [I] have also characterized a kidney tubular antigen with nephritogenic activity: McPhaul and D ixon [26] have isolated a glomerular basement membrane antigen in normal adult serum and urine. Even with all this knowledge, the behavior of most of the kidney tissue antigens is still obscure.…”

“…Previously, D inh [7] had prepared a 'major antigen' of rat kidney after ultracentrifugation of renal tissue homogenates, followed by chromatography on DEAE-cellulose and gel filtration on Sephadex G-200; Srinivasan et al [8] have extracted cortex proteins by precipitation with an ammonium sulphate solution saturated between 60 and 100%.…”
