Isolation and characterization of Staphylococcus aureus starvat ion4 nduced, stationary-phase mutants defective in survival or recovery

Abstract: Ten Staphylococcus aureus mutants, defective in the starvation-induced stationary phase of growth were isolated from two independent Tn917-LTVI transposon insertion libraries and were designated suv as they had apparent -suryival defects. Seven of these mutants were defective under amino-acidlimiting conditions alone. Two mutants (suv-3 and suw-20) demonstrated lower plating efficiency when starved for glucose, phosphate or amino acids and one mutant (suv-11) had reduced plating efficiency after amino acid or … Show more

“…The other mutants lacked SodA, which has been discussed above; RpoE, which seems to play a regulatory role in stationary phase; a protein of unknown function; and UmuC. UmuC plays a role in the SOS response in B. subtilis, i.e., the response induced in response to DNA damage (242), again demonstrating the importance of DNA repair systems in the response to low pH (245).…”

“…Starvation survival experiments were carried out in glucose-limiting CDM medium, with shaking at 250 rpm at 37°C (55). Desiccation tolerance testing was performed on stationary-phase cells grown in CT-BHI, washed, and resuspended in PBS to an OD 600 of 0.05.…”

“…To measure growth in the presence of high-salt concentrations, cultures were inoculated from overnight BHI cultures to an OD 600 value of 0?08 into either BHI or BHI containing 2 M NaCl at 37 uC. Hydrogen peroxide resistance assays were carried out as described previously (Watson et al, 1998a) with the following modifications. Cells were grown in amino-acid-limiting CDM (1 %, w/v, glucose) to exponential phase (OD 600 0?1).…”

“…The Tn917 insertion mutant SPW20 was isolated in a previous screen for starvation survival mutants of S. aureus (Watson et al, 1998a). In this strain, the transposon is inserted in an ORF of 185 aa that shares 59 % identity with the putative hypoxanthine-guanine phosphoribosyltransferase (HprT) enzyme from B. subtilis.…”
