Pathogens form biofilms to increase their resistance to environmental stress and antibacterial compounds. The rhizosphere is a rich source of microorganisms producing industrially important compounds including those with antimicrobial and biofilm inhibitory activities. Four isolates from soil collected from Taguig City, Philippines, were subjected to phenotypic and genotypic characterization, screening for protease production, and biofilm inhibition assays. Colony morphology and microscopic analyses indicated the isolates were putative Bacillus species. Upon DNA extraction, 16S rRNA gene was amplified, and based on their sequences, the isolates were confirmed to be Bacillus spp. Isolate AHP was B. cereus, isolate DJA was Priestia megaterium, formerly known as B. megaterium and isolates SJS and SKA were Bacillus spp.-all of which produced protease. Although the cell-free supernatants (CFS) of the isolates did not inhibit the growth of Staphylococcus aureus 1258, Citrobacter freundii ATCC24864, Salmonella Typhimurium ATCC13311, Escherichia coli ATCC11229, and E. coli O157:H7, biofilm formation of S. aureus was inhibited by all CFS, with B. cereus AHP showing the highest biofilm inhibition at 46%, followed by Bacillus sp. SKA (39%), P. megaterium DJA (36%), and Bacillus sp. SJS (31%). Even though further studies are warranted, the bioactivities of these isolates indicate potential use for pharmaceutical purposes due to their ability to produce protease and inhibition of biofilm formation of a common bacterial pathogen.

Research about the isolation, screening and partial characterization of thermophilic bacteria that produce protease from Bukik Gadang hot springs, Solok Regency, was carried out at the Basic Biology Laboratory, UPT Basic and Central Laboratory, Andalas University in January - April 2022. This study aims to obtain isolates of thermophilic bacteria that indicated protease production and to determine the partial characterization of thermophilic bacteria that indicated protease production from Bukik Gadang hot springs, Solok Regency. This study used a survey method and purposive sampling technique. The results of this study obtained 18 isolates from 20 isolates of thermophilic bacteria which indicated protease production, with one isolate of TPBG-03 having the potential to produce protease. Partial characterization by 10 isolates of thermophilic bacteria indicated protease production was generally Gram positive, bacillus cell shape, spore-forming, catalase positive and motile.Keywords - Hot Springs, Isolation, Protease, Screening, Thermophilic Bacteria

Forty soil samples were collected from different regions in Baghdad city. Among 52 isolate, only 38 isolates (73.1 %) belong to Bacillus spp. All bacterial isolates have been submitted to the cultural, microscopical, biochemical examination and VITEK 2 System, Bacillus cereus was predominantly found 21 (55.3%) followed by Bacillus subtilis 11 (28.9%), then Bacillus amyloliquefaciens was 6 (15.8%). Bacillus cereus (B1) and Bacillus subtilis (B16) isolates were with the highest level with the antibacterial activity when selected. Extracellular proteins were extracted and purified with ammonium sulfate (80%) saturation, then submitted to Sepharose-6B gel filtration column. SDS-polyacrylamide gel electrophoresis was used to determine the molecular weights of proteins, and one band appeared to each of Bacillus cereus and Bacillus subtilis with molecular weight 68 and 50 KDa respectively. The results revealed that higher effect of antibacterial activity on Gram positive bacteria, and moderate on Gram negative bacteria except P.aeruginosa with no effect. The antibiofilm effect of the extract found higher effective against S.pyogens, moderate effect against each of (S.aureus, E.coli, S. marcescens, K. spp) and no effect against P.aeruginosa. The results of cytotoxicity showed that the purified extract was a higher cytotoxic effect on breast cancer MCF7 cells at 1000µg/ml concentration, while the crude extract had a higher cytotoxic effect on breast cancer MCF7 cells at 100µg/ml concentration.
