Isolation and characterization of phosphate solubilizing bacteria naturally colonizing legumes rhizosphere in Morocco

Abstract: Low-cost and environmentally friendly agricultural practices have received increasing attention in recent years. Developing microbial inoculants containing phosphate (P) solubilizing bacteria (PSB) represents an emerging biological solution to improve rhizosphere P availability. The present study aims to explore PSB strains isolated from soils located at different bioclimatic stages in Morocco and present in various legumes rhizosphere to improve agronomic microbial fertilizer’s effectiveness. It was also aime… Show more

“…Plant nutrient models have long demonstrated that slow diffusion of inorganic phosphate (Pi) is a major barrier to P acquisition by plants. Soil P is characterized by its restricted mobility when compared to other important nutrients, such as nitrogen or potassium Janati et al (2022). In the present study, Pikovskaya's media, which utilizes tricalcium phosphate as the only source of phosphorus, was used for the initial screening of the phosphate-solubilizing bacterial strain.…”

“…The available P portion was determined by a spectrophotometer at 880 nm using the ascorbic acid colourimetric method. The absorbance was measured using standard solutions of KH 2 PO 4 with P concentrations ranging from 0.1 to 1 mg.L −1 ( Janati et al, 2022 ).…”

“…Phosphate solubilizing bacteria were isolated on National Botanical Research in P medium (NBRIP) agar plates [18,19]. To determine the efficiency of P solubilization, the PSB isolates were grown in an NBRIP agar medium supplemented with 5 g.L -1 TCP.…”

“…A standard range of KH 2 PO 4 (concentration from 0 to 1 mg.L -1 ) was used to calculate P content. To identify PSB genera, we extended the morphological and biochemical tests conducted by Janati et al (2022) [19]. In this paper, we examine the preliminary screening of PSB isolates and investigate the species affiliation of each potential isolate for phenotypic characterization of phosphate solubilizing bacteria based on growth in a different medium, according to Bergey's textbook (Systematic Bacteriology, 2nd ed., vol.5, eds., 2012).…”
