Isolation and characterization of permanent cell lines from inner cell mass cells of bovine blastocysts

Abstract: Inner cell masses (ICM) from in vitro produced day 8 or 9 bovine blastocysts were isolated by immunosurgery and cultured under different conditions in order to establish which of two feeder cell types and culture media were most efficient in supporting attachment and outgrowth of the bovine ICM cells. The efficiency of attachment and outgrowth of the ICM cells could be markedly improved when STO feeder cells were used instead of bovine uterus epithelial cells, and by using charcoal-stripped serum instead of no… Show more

“…There are several reports about the establishment of bovine ES-like cells from ICM cells [5][6][7][8] and birth of calves after nuclear transfer of these cells [6]. From the results obtained in the present study, if bovine ES-like cells form a clump to some extent after the trypsinization like mammary cells, they would contribute to ICMs of aggregates as well as in mice.…”

“…Furthermore there have been several reports on the establishment of bovine ES cell-like cell lines [5][6][7][8]. Therefore there are some possibilities to produce cloned calves efficiently, if bovine ES cell-like cells can be treated with this aggregation technique using tetraploid embryos.…”

“…This result suggests that an increased glucose concentration, instead of increasing glucose metabolism, is detrimental to 5-day-old embryos (morula). Although the role of GH on embryo development is not clear, GH was reported to increase the glucose uptake of the mouse embryo [19]. This effect of GH on glucose transport is due to direct recruitment of the transporters GLUT 1 and GLUT 4 to the plasma membrane [20].…”

“…Thomson's group [6] derived human ESC lines from embryos with normal karyotypes and the potential to develop into trophoblasts and three embryonic germ layers. Progress has also been made in other species, including pigs [7][8][9], cattle [7,[10][11][12][13], sheep [14,15], rabbits [16], horses [17], dogs [18], and cats [19], but these studies did not fulfill all characteristics of mESCs. Also, at this time, researchers have failed to derive genuine ESCs from domesticated ungulates such as pigs, cattle, goats, sheep and derivation of germline transmittable chimeric offspring has proven to be elusive in these species.…”
