Isolation and Characterization of Human Natural Killer Cell Subsets

Abstract: Natural killer (NK) cells are innate immune lymphocytes that play a critical role in the host defense against pathogens through their production of cytokines and cytotoxic activity. Human NK cells can be divided into two subsets, each with distinct phenotypic and functional properties, based on cell-surface density expression of CD56 (CD56bright and CD56dim). This unit describes the purification of human NK cell subsets from blood, protocols for the further characterization of NK cell function, and further bac… Show more

“…Normally, NK cells in the peripheral blood of healthy subjects have the phenotype 90% CD16 + and 10% CD16 - . The CD16 + phenotype is associated with activation and cytotoxicity, while the CD16 - cells release cytokines and are not cytotoxic [102]. In the PF of EOC patients, however, this ratio shifts to 60% CD16 + and 40% CD16 - .…”

“…In general, cytotoxic activity is associated with CD56 Dim pNKs and cytokine production with CD56 Bright pNKs(12, 13). However, CD56 Dim pNK cells are also capable of cytokine production and CD56 Bright pNKs may acquire cytotoxic capacity upon cytokine induced activation(14-16). …”

“…These soluble forms are increased in a variety of tumors and may serve as a tumor evasion mechanism [106] . According to the frequency of CD56 expression (dim or bright) and the presence or absence of CD16, NK cells can be divided into two subsets: CD56 bright CD16 neg (produces cytokines) and CD56 dim CD16 pos (cytotoxic) [107] . The density of NK cells in the peripheral blood of patients with HCC positively correlated with survival and prognosis similar to many of the other cell lines already mentioned [108] .…”
