Isolation and characterization of a Pseudomonas aeruginosa bacteriophage with a very limited host range

Abstract: A Pseudomonas aeruginosa bacteriophage, phi PLS743, with extremely limited host range has been isolated. It belongs to the virus family Podoviridae, morphological type C1, and possesses a head diameter of 45 nm. The phage has a buoyant density in CsCl of 1.516 g/cm3, and its mass is 45 x 10(6) daltons. The phage particles are composed of double-stranded DNA (49.9 mol% G + C; 42.4 kilobase pairs) and 11 structural proteins (66% by weight). The major head protein, P5, has a Mr of 34,500. The DNA is not cut by Sa… Show more

“…Head and tail dimensions of these phages closely resembled with some of the earlier reported Vibriophages such as N1, N2, N3, N4, N5 and S5 belonging to Podoviridae family [44,25]. The findings of this study also support the observations made by earlier workers where phages such as ФPLS743, LK16, LKA1 and ФKMV against Pseudomonas aeruginosa belonging to Podoviridae family were isolated [45][46][47]. To the best of our knowledge, this is the first report on isolation and characterization of bacteriophages specific to Pseudomonas aeruginosa PAO, belonging to family Podoviridae from India.…”

“…Although phages infecting strains of two or more species of a particular genus have been described, the results to date indicate that most of them are host species-specific and also demonstrate strain specificity [4,8,10]. All of the isolated phages in this experiment infected only Xoo strains.…”

“…Among the strains tested it lysed only P. fluorescens R12t, which is very similar to strain R2f. This specificity is not surprising because many bacteriophages have a limited host range, which in some cases is used for strain typing (1,3). Therefore, this phage was considered to be sufficiently specific to selectively kill off the introduced donor cells, leaving undisturbed the majority of the indigenous soil bacteria.…”
