“…This study shows that acetylcholine and muscarine stimulate the 32Pi incorporation into phosphatidic acid, the efflux of 45Ca2+, and the synthesis of cyclic GMP in cultured bovine chromaffin cells. Our observation is consistent with other reports [3,14-171 that acetylcholine stimulated the turnover of phosphatidylinositol in a variety of tissues via muscarinic receptor, and the reports [5, 15,16] that muscarinic agents stimulated the efflux of 45Ca2+ and increased cyclic GMP levels in pancreatic acinar cells, pituitary glands and chromaffin cells. We further demonstrate that phosphatidic acid exogenously added to cultured chromaffin cells mimicks the effect of acetylcholine in that it induces the efflux of 45Ca2+ from prelabelled cells and elevates the intracellular cyclic GMP content, thus extending our recent finding on cloned neuroblastoma NlE 115 cells [4].…”

“…Cell preparation and culture Bovine adrenal chromaffin cells were dispersed enzymat ically as described previously (15). The cells were plated in 35-mm culture dishes at a density of 2 x 106 cells/dish for standard experiments or on 22 x 22 mm cover glasses in 35-mm culture dishes at a density of 1 x 106 cells/dish for measuring intracellular calcium.…”

“…Physiological stimulation such as acetylcholine for adrenal chroma n cells increases the level of [Ca 2+ ] i originated from intracellular pools and extracellular spaces, leading to biosynthesis and secretion of catecholamine in the cells (Oka et al, 1979;Masserano et al, 1990). The adrenal medulla is rich containing vascular endothelial cells and chroma n cells.…”

“…Bovine adrenal chroma n cells were dispersed enzymatically as described previously (Oka et al, 1979). Brie¯y, the medulla was sliced with a hand slicer, and the slices were digested in medium containing 0.1% collagenase, 0.01% soybean trypsin inhibitor and 0.5% bovine serum albumin in balanced salt solution [BSS: 135 mM NaCl, 5.6 mM KCl, 1.2 mM MgSO 4 , 2.2 mM CaCl 2 , 10 mM glucose, and 20 mM N-2-hydroxyethylpiperazine-N'-2-ethanesulphonic acid (HEPES)/NaOH, pH 7.4].…”
