Isoform Specificity in the Relationship of Actin to Dendritic Spines

Abstract: Dendritic spines contain high concentrations of actin, but neither the isoforms involved nor the mechanism of accumulation is known. In situ hybridization with specific probes established that beta- and gamma-cytoplasmic actins are selectively expressed at high levels by spine-bearing neurons. Transfecting cultured hippocampal neurons with epitope-tagged actin isoforms showed that cytoplasmic beta- and gamma-cytoplasmic actins are correctly targeted to spines, whereas alpha-cardiac muscle actin, which is norma… Show more

“…For all experiments in neurons, a vector using the chicken ␤-actin promoter (pBetaActin) was used to drive expression (Kaech et al, 1997). Plasmids encoding WAVE-1 were generated by PCR amplification of DNA and ligation of the restriction digested insert at sites downstream of enhanced green fluorescent protein (GFP) (Clontech).…”

“…Actin remodeling underlies the establishment and maturation of dendritic spines, and ␤-actinimmunoreactive levels are a measure of dendritic spine density (Matus et al, 1982;Kaech et al, 1997). To begin analyzing the mechanism of estradiol action, phosphorylated cofilin was measured (Schubert et al, 2006;Carlisle et al, 2008).…”

“…A viral epitope such as the 11-amino acid peptide derived from vesicular stomatitis virus G protein (VSV-G) (Soldati and Perriard, 1991) decreases the risk that the antibody recognizing the tag crossreacts with cellular components. Insertion of this particular tag at the C terminus of different actin isoforms has been used to study their distribution relative to the endogenous actins in fibroblasts and cardiomyocytes (von Arx et al, 1995), smooth muscle, endothelial and epithelial cells (Mounier et al, 1997), and hippocampal neurons (Kaech et al, 1997). For the interpretation of these experiments it has been assumed that the tag does not interfere with the correct folding and function of the protein.…”
