Isoform‐specific expression and characterization of 14‐3‐3 proteins in human glioma tissues discovered by stable isotope labeling with amino acids in cell culture‐based proteomic analysis

Abstract: Human 14-3-3 proteins have isoform-specific expression and functions in different kinds of normal or tumor cells and tissues. However, the expression profiling of 14-3-3 proteins and isoform-specific biological functions are unclear in human glioma so far. In our study, the expression levels and characterization of 14-3-3 isoforms in human glioma tissues were investigated by a sensitive, accurate stable isotope labeling with amino acids in cell culture-based quantitative proteomic strategy. As a result, except… Show more

“…The effect of 14-3-3 proteins on cancer development and their interaction with various oncogenes and tumor suppressor genes is complex (36). A previous study investigated the increased immune reactivity of various 14-3-3 isoforms in human astrocytoma samples, and the immunoreactivity of cells was significantly enhanced with increased disease progression (37).…”

“…10 In addition, 14-3-3␤ and 14-3-3 were found specifically expressed in human astrocytoma. 11,12 The expression frequency and levels of 14-3-3␤ and 14-3-3 were associated with human astrocytoma. 12 Moreover, increased expression of 14-3-3 in breast cancer was shown in a proteomic study.…”

“…Роль белков 14-3-3 в злокачественных опухолях и их взаимодействие с различным опухолеродными генами и генами-супрессорами опухоли сложна и ма-ло изучена [32]. Предыдущие исследования продемон-стрировали, что иммунореактивность изоформ 14-3-3b, 3, d, q, h и z наблюдалась в большинстве образцов с астро-цитом, и их иммунореактивные показатели заметно увеличивались с ростом стадии заболевания [33].…”
