Species of Nicotiana grow naturally in different parts of the world and have long been used both medicinally and recreationally by human societies. More recently in our history, Nicotiana tabacum has attracted interest as one of the most economically important industrial crops. Nicotiana species are frequently investigated for their bioactive natural products, and the ecological role of their specialized metabolites in responses to abiotic stress or biotic stress factors like pathogens and herbivores. The interest of tobacco companies in genetic information as well as the success of a few wild tobacco species as experimental model organisms have resulted in growing knowledge about the molecular biology and ecology of these plants and functional studies of the plant's natural products. Although a large number of reviews and books on biologically active natural products already exists, mostly from N. tabacum, we focus our attention on the ecological roles and biological activity of natural products, versus products from cured and processed material, in this Review. The studied compounds include alkaloids, aromatic compounds, flavonoids, volatiles, sesquiterpenoids, diterpenes alcohols, and sugar esters from trichomes of the plants, and recently characterized acyclic hydroxygeranyllinalool diterpene glycosides (HGL-DTGs). In this Review (1800s-2017), we describe the above-mentioned classes of natural products, emphasizing their biological activities and functions as they have been determined either in bioassay-guided purification approaches or in bioassays with plants in which the expression of specific biosynthetic genes has been genetically manipulated. Additionally, a review on the history, taxonomy, ecology, and medicinal application of different Nicotiana species growing around the globe presented in this Review may be of interest for pharmacognosists, natural products, and ecological chemists.

Background: All over the world, natural products containing different secondary metabolites have been used for antibacterial purposes, and as folk medicines with significant effects. Amongst many different plants, Tobacco plants are cultivated all over the world, but natively belong to America. These plants contain variety of secondary metabolites and possess significant antibacterial activity. Objectives: To conduct phytochemical analysis and measure the antibacterial potential of Tobacco plants Nicotiana tabacum and Nicotiana rustica using their extracts. Methodology: Multiple chemical tests such as Wagner’s test and Fehling’s test were used to determine the presence of different types of secondary metabolites. Both plant species were also screened for their antibacterial activity using agar well diffusion method. Results: Phytochemical analysis of the extracts from both plant species i.e. Nicotiana tabacum and Nicotiana rustica indicated the presence of secondary metabolites including tannins, alkaloids, terpenoids, saponins, steroids, and flavonoids. Significant antibacterial activity of both plant extracts was observed against Staphylococcus aureus, but not against Escherichia coli. Conclusion: It can be concluded that both plant extracts showed the presence of secondary metabolites, with significant inhibitory effect observed against Staphylococcus aureus, and no effect against Escherichia coli.

Under natural conditions, the plants are, usually, surrounded by a series of potential enemies. They had created strategies of defense against pathogens and herbivores' attacks, allowing its perpetuation throughout evolution. These defense mechanisms are closely associated with the synthesis of secondary metabolites, which are also worldwide used in several areas of industry. This bibliographic review was conducted aiming to better understand how plants synthesize these substances in response to biotic or abiotic stimuli. The results discussed here revealed that synthesis of plant secondary metabolites is dependent on changes in several metabolic pathways, being often directly associated with the primary metabolism. Injury by herbivores or pathogens, temperature, CO 2 levels, solar radiation and drought, are the factors related to the environment that express the most significant signs of inhibition or synthesis of plant secondary metabolites. Global climatic changes recently observed can affect the metabolic pathways network and, consequently, secondary metabolites synthesis. Nowadays, genomic tools have been useful alternatives that are leading to a new revolution of plant breeding, allowing the overexpression or inhibition of these substances. Some limitations and challenges to be achieved upon the dynamics of secondary metabolite synthesis in plants are presented.
