Ischaemia and reperfusion injury of rat liver increases expression of glutathione S-transferase A1/A2 in zone 3 of the hepatic lobule

Abstract: Effects of ischaemia-reperfusion injury (I\R) of liver on expression of rat glutathione S-transferase (rGST) isoenzymes that metabolize products of oxidative stress were examined. Rats underwent lobar liver ischaemia for 30 min followed by reperfusion. In ischaemic lobes, rGSTA1\A2 transcript levels increased significantly 12 h after I\R (2.94-fold) and protein levels increased significantly at 24 h (1.45-fold) ; increased transcript levels were also observed in nonischaemic lobes (1.78-fold). Superoxide dismu… Show more

“…Oxidative stress, due either to drugs or to events such as ischemia-reperfusion injury, increases expression of alpha class genes in mouse and rat livers.l,6,66 Induction by drugs, such as phenobarbital, and that following ischemia and reperfusion injury is limited to zones 2 and 3 of the hepatic lobule, whereas zone 1 hepatocytes are unaffected. 66 The GST genes are present in all hepatocytes, and the lobular heterogeneity appears to reflect differences in induction pathways in the different hepatocytes.…”

“…The level of expression of GSTs and their biochemical properties are crucial factors in determining cellular resistance to carcinogens, antitumor drugs, environmental pollutants, and products of oxidative stress (2)(3)(4)(5). All eukaryotes possess multiple GST isoenzymes, each displaying distinct catalytic as well as noncatalytic binding properties (6,7).…”

“…The area observed in Teramoto's report was from the central venous area to the portal area. However, Rappaport et al 28 and Branum et al 29 reported that zone 3 of the liver was sensitive to ischemic damage. Therefore, we measured the zone 3 SD together with the Vv modified from Miller's parameter.…”
