“…In fact, sphingosine also acts as an activator of DAG kinase, and it has been shown that DAG produced from PIP2 may not be a better substrate for DAG kinase activated by sphingosine (44). R59 949 is a very specific and potent DAG kinase inhibitor (45) and has been shown to inhibit DAG kinase in a concentration dependent manner (0.01 to 10 ~tM) in isolated platelet membranes (IC50=0,3 ~M) and in intact cells (ICs0=0.12 t~M). The fact that DAG kinase inhibitor R59 949 did not completely abolish the PA formation by sphingosine suggests that PLD is also activated by sphingosine apart from that mediated by PC-PLC and DAG kinase.…”
