Is There Any Relationship between Human Herpesvirus-8 and Multiple Myeloma?

Abstract: Background. Human herpesvirus-8 (HHV-8) is associated with some human diseases including Kaposi's sarcoma and also some B-cell lymphoproliferative disorders. Few studies have highlighted the potential role of HHV-8 in the development of multiple myeloma (MM) which is known as a malignant proliferation of plasma cells derived from a single clone. Aims. The aim of this study was to find a relationship between HHV-8 and MM using polymerase chain reaction (PCR) method. Materials and Methods. This study was conduct… Show more

“…Rettig et al described the presence of Kaposi’s sarcoma associated herpesvirus infection in bone marrow dendritic stromal cells in 1992 with the postulation that the viral homologue of interleukin 6 coded for by HHV8 might drive the progression of monoclonal gammopathy of uncertain significance to overt myeloma through paracrine stimulation 4. This hypothesis has been challenged and there have been several conflicting bodies of evidence from studies conducted in different geographical areas 4 5. This might stem from different methodologies used and also the diverse epidemiology and seroprevalence rates of HHV8 across different ethnicities and geographical locations.…”

“…Relationship between viruses and pathogenesis of various benign and malignant tumors remain a concern for physicians [22][23][24][25]. JAK/STAT signaling pathway is one of the most important cytokine-signaling pathways regulating T-lymphocyte function.…”

“…Spinal cord compression following vertebral compression fractures or vertebral plasmacytoma comprises 5% of the presentation of multiple myeloma [7]. The aetiology of multiple myelomais unknown but some studies have suggested the pivotal role of viruses in the development of multiple myeloma [15]. Diagnosis of multiple myeloma involves demonstrating the monoclonal immunoglobulins in the serum or urine, demonstration of more than 10% of malignant plasma cells in the bone marrow with or without the end organ effects like anaemia, hypercalcemia or renal impairment.…”
